In vivo ādas fluoroforu raksturošana ar fluorescences dzīveslaika spektroskopiju

Darba mērķis ir raksturot cilvēka in vivo ādā esošās fluorescentās komponentes – fluoroforu – kā parametru izmantojot to fluorescences dzīves laiku; noteikt nepārtrauktas darbības zemas jaudas 405 nm lāzera starojuma ierosinātas fotoizbalēšanas ietekmi uz fluorescences dzīves laiku.
Darba gaitā veikta fluoroforu sadalījuma un attiecīgo fluorescences dzīves laiku novērtēšana no 400 ādas apgabaliem un iegūta stabila literatūrai atbilstošs vidējo vērtību sadalījumus I – III fototipa ādai. Modeļa eksperimentā un ādas paraugiem testēta no laika atkarīgā emisijas spektra uzņemšana (TRES) un noteikta fluorescences signāla evolūcija laikā. Spektra reģionā no 560 - 680 nm tika notiekts, ka flavīns nes lielāko ieguldījumu ādas autofluorescences spektra radīšanā.
Konstatēta fluorescences dzīves laiku samazināšanas fotozibalēšanas procesā. Pēc 6 min fotoizbalēšanas datu sakritība ar teorētisko modeli ievērojami uzlabojas.

Work includes finding how stabile the average value of skin autofuorescence lifetimes is amongst different subjects. It includes fluorescence lifetime measurements from 400 skin regions amongst skin prototypes I to III. Model experiments and in vivo skin experiments using time resolved emission spectroscopy (TRES) method were carried out and evolution in time of the fluorescence signal were determined. Flavin was found as main fluorescent component of skin autofluorescence spectrum region 560 - 680 nm.
Study shows that photobleaching effect has an effect on measured fluorescence lifetime, after 6 min of photobleaching lifetime components decrease and data shows better fit to theoretical model.
