Superoksīda un NO daudzuma noteikšana aortā un asinīs

Aterosklerozes attīstība saistīta ar paaugstinātu superoksīda un samazinātu slāpekļa oksīda (NO) sintēzi asinsvadu endotēlijā. Bakalaura darbs veltīts superoksīda un NO kvantificēšanai apolipoproteīna E „knockout” (ApoE-/-) peļu, kas ir plaši izmantots eksperimentālās aterosklerozes modelis, un ICR peļu aortās un asinīs, kā arī NO sintēzes novērtēšanai žurku aortās, izmantojot diaminofluorescīna diacetāts(DAF-DA).
No iegūtajiem rezultātiem varam secināt, ka apoE-/- pelēm salīdzinot ar ICR pelēm bija trīs reizes zemāks nitrozilhemoglobīna (NOHb) daudzums asinīs. ApoE-/- peļu aortās atšķirībā no ICR peļu aortām tika konstatēta izteikta etīdija bromīda fluorescence, kas liecināja par aktīvu superoksīda producēšanos endotēlijā un aortas muskulatūrā. Karbamoilholīns izraisīja DAF-DA fluorescenci, kas apstiprināja NO izdalīšanos žurku aortās.

The development of atherosclerosis is related to elevated superoxide and decreased nitric oxide (NO) synthesis in the vascular endothelium. The bachelor thesis is devoted to quantifying superoxide and NO in the aortas and blood of apolipoprotein E „knockout” (ApoE-/-) mice, which is widely used model of experimental atherosclerosis, and ICR mice, as well as assessing the NO synthesis in the aortas of rats using diaminofluorescein diacetate (DAF-DA).
Obtained data demonstrated that the level of nitrosylhemoglobin in the blood of apoE-/- mice compared to ICR mice was three times lower. In the aortas of apoE-/- mice unlike those of ICR mice, distinct presence of ethidium bromide fluorescence was found which indicated an active production of superoxide in the endothelium and smooth muscle cells. Carbamoylcholine induced fluorescence of DAF-DA which proved the synthesis of NO in the aortas of rat.
