Vēzis ir viena no mūsdienu medicīnas svarīgākajām problēmām. Mērķēta alfa-terapija, kas izmanto pie bioloģiskiem nesējiem piesaistītus α-daļiņas emitējošus radionuklīdus un tiek mērķēta selektīvi uz malignajām šūnām, ir veids, kā iznīcināt atsevišķas vēža šūnas, pat ja tās ir izkaisītas pa visu organismu.
Šī darba mērķis ir apkopot informāciju par α-emiteru izmantošanu onkoterapijā, kā arī akcentēt galvenos farmācijas un ķīmijas saskares punktus uz audzēja šūnām mērķētu radiofarmaceitisko vielu izstrādē.
Darba teorētiskajā daļā apkopota zinātniskajā literatūrā pieejamā informācija par α emiteriem, to pieejamību, darbības principu, priekšrocībām un potenciālajām iespējām onkoterapijā. Sniegts vispārīgs priekšstats par mērķētu pretvēža terapiju, tajā lietotajiem bioloģiskajiem pārneses moduļiem, kā arī aplūkoti biežāk izmantotie helatori radionuklīdu kompleksēšanai. Literatūras apskats aptver laika periodu no 1993. līdz 2015. gadam, izmantotas PubMed un Journals of American Chemical Society datu bāzes.
Darba praktiskajā daļā sintezēts jauns 227Th kompleksēt spējīgs savienojums, kas satur terminālo dubultsaiti un tādējādi ir pakļaujams tālākajām modifikācijām, lai to varētu pievienot pie bioloģiskas nesējmolekulas. Sākotnējie mēģinājumi sintezēt molekulu, kas satur brīvu karboksilgrupu, bija neveiksmīgi, jo visos gadījumos radās nevēlams blakusprodukts – laktons. Arī molekulu, kura saturētu benzilspirta grupu sintezēt neizdevās, jo anhimērā blakusgrupas efekta dēļ notika vēlamā savienojuma debenzilēšanās un benzilgrupas migrācija uz vienu no izejvielām. Cancer is one of most important problems of modern medicine. Targeted alpha-therapy using α-particles emitting radionuclide bound to biological carrier which is targeted selectively to the malign cells is one of the way how to destroy single cancer cells even if they are spread throughout the body.
The objective of the bachelor’s thesis is to summarize the literature data about the therapeutic use of α-emitters as well as to highlight the main contact points of pharmacy and chemistry in the development of tumor targeted radio-pharmaceuticals.
The theoretical part of the thesis includes the scientific literature review about the α emitters, their availability, mechanism of action, benefits and potential use in tumor therapy. The review also provides insight into the tumor targeted therapy, most recently used biological carriers and chelate compounds capable of complexing a radionuclides. Publications from year 1993 to 2015 from PubMed and Journals of American Chemical Society data bases were used.
In the practical part a new 227Th complexing compound has been synthesized. The molecule has terminal double bond which could be modified further in order to be attached to the biological carrier. Initial attempts to synthesize a molecule containing free carboxyl group failed since an undesired by-product – lactone – formed in all cases. Also the synthesis of an alternative molecule containing benzylalcohol group was not successful due to anchimerically assisted debenzylation of the formed compound and migration of the benzyl group to one of the starting materials.
