A. Kazāka vadībā. This study reports sequence analysis of bacteriophage Enc34 replication module proteins and experimental work on hypothetical proteins ORF3, ORF4, ORF5 and ORF6 to identify their function. During the study, a previously undetected helicase domain was found in the primase ORF1, and ORF8 was reannotated as resolvase. Recombinant ORF4 was insoluble, while ORF3 and ORF5 were soluble and successfully purified, allowing to proceed with their crystallization. Crystal structures of ORF6 and a deletion variant ORF6Δ21 were determined, which together with in vitro assays showed that ORF6 is a ssDNA-binding protein (SSB). This is the first study that demonstrates the role of the SSB C-terminal region for ssDNA binding specificity. The work was conducted at the Latvian Biomedical Research and Study Centre under the supervision of Dr. biol. A. Kazāks.
