Ģenētiskās izmaiņas pārmantotās melanomas pacientiem Latvijā

Melanoma ir multifaktoriāla slimība, kas attīstās ādas pigmenta šūnu, melanocītu, nekontrolētas dalīšanās rezultātā. Bakalaura darba mērķis bija izvērtēt izmaiņas augsta un vidēja riska melanomas jutības gēnos pacientiem, kuru anamnēzē ir vairāki saslimšanas gadījumi ar melanomu vai arī saslimšanas gadījumi ar aizkuņģa dziedzera audzēju vai nervu sistēmas audzēju. Kopā darbā izanalizēti 39 melanomas pacienti, 9 pacientu ģimenes locekļi un viens pacients ar acs melanomu. Ar paralēlo daudzķēžu sekvenēšanas metodi izanalizētajos 4 gēnos (BAP1, POT1, CDKN2A un MC1R) kopumā atrastas 11 nomaiņas. Papildus 3 analizētajos gēnos (TERT, CDK4 un MITF) izmaiņas atrastas netika. Kā potenciāli bīstamas novērtētas līdz šim neaprakstītas nomaiņas BAP1 (Pro293Leu) un POT1 (Leu153STOP). Atslēgas vārdi: melanoma, riska gēni, HaloPlex™ platforma, nomaiņas. Melanoma is a multifactorial disease which develops from uncontrolled division of melanocytes. The purpose of the bachelor’s work was to analyse alterations in high risk and low risk melanoma susceptibility genes of melanoma patients with multiple melanoma cases or pancreatic or nervous system cancer cases in their anamnesis. A total of 39 melanoma patients, 9 of their relatives and one patient with uveal melanoma were analysed. With the parallel multi-chain sequencing method 11 SNPs were found in 4 genes: BAP1, POT1, CDKN2A and MC1R. No alterations were found in three genes: TERT, CDK4 and MITF. Two of the SNPs were evaluated as potentially harmful - Pro293Leu in BAP1 and Leu153STOP in POT1 - and haven’t been previously described.
