Legionella pneumophila izplatība un sekvenču tipu daudzveidība Latvijas viesnīcu ūdensapgādes sistēmās

Legionella pneumophila ir nosacīti patogēna, dabiskās un mākslīgi radītās saldūdens ūdenskrātuvēs sastopama baktērija, kas spēj izraisīt infekcijas slimību – legionelozi. Lai noskaidrotu L. pneumophila izplatību Latvijā, no 40 viesnīcu ūdensapgādes sistēmām noņemti 240 karstā ūdens paraugi un testēti uz L. pneumophila klātbūtni. No pētījumā iesaistītajām viesnīcām 25 (65 %) viesnīcās konstatēts vismaz viens Legionella pozitīvs paraugs. Maksimālais L. pneumophila koloniju veidojošo vienību skaits vienā paraugā bija 1,1 x 104 kvv/l. Ar MALDI-TOF MS apstiprināti 99 L. pneumophila un 9 L. rubrilucens izolāti. Pārstāvētākā no serogrupām bija L. pneumophila serogrupa 3 (68 izolāti). Izmantojot SBT metodi iegūti 26 sekvenču tipi, no kuriem četri (ST 2579, ST 2580, ST 2581 un ST 2582) EWGLI datubāzē reģistrēti pirmo reizi. Visvairāk izolātu identificēts ar sekvenču tipa numuru ST 1104 (15 izolāti). Iegūta minimum-spaning tree dendrogramma, kurā 26 sekvenču tipi iedalās 4 grupās (A,B,C un D) un 11 atsevišķos “singletons” sekvenču tipos. Šis pētījums papildina EWGLI datubāzē iekļauto L. pneumophila sekvenču tipu skaitu, palīdz iegūt informāciju par to ģenētisko dažādību Latvijā un pasaulē, kā arī saskatīt L. pneumophila radniecību starp atšķirīgajiem sekvenču tipiem. Legionella pneumophila is a conditionally pathogenic bacteria found in natural and man-made freshwater reservoirs which can cause infectious disease – legionellosis. To find out the prevalence of L. pneumophila in Latvia, total of 240 hot water samples from 40 hotel water supply systems were taken. Overall, at least one positive sample of Legionella was found in 25 (65 %) of study involved hotels. The maximum number of L. pneumophila colony forming units in a sample was 1.1 x 104 cfu/l. In total, 99 L. pneumophila and 9 L. rubrilucens isolates were identified by MALDI-TOF MS. The most frequently L. pneumophila serogroup was serogroup 3 (68 isolates). In this study was used SBT method, which all isolates were classified into 26 sequence types, including four new sequence types (ST 2579, ST 2580, ST 2581 and ST 2582) that were submitted in EWGLI database for the first time. A minimum-spanning tree dendrogram was obtained from 26 sequence types containing 4 groups (A, B, C and D) and 11 singleton sequence types. This study complements the EWGLI database with new L. pneumophila sequence types, demonstrated genetic diversity not only in Latvia but also in the world, as well as showed relationship between different L. pneumophila sequence types.
