Elpošanas ķēdes loma piruvātu producējošos Zymomonas mobilis celmos

Lai novirzītu Zymomonas mobilis katabolismu no etanola producēšanas uz piruvāta producēšanu, jāatrod veids, kā oksidēt to NADH, kas parasti tiek izmantots etanola sintēzei, bet nav nepieciešams piruvāta producēšanai. Elpošana ir viens veidiem, kā oksidēt NADH. Lai gūtu priekšstatu par elpošanas ķēdes ietekmi uz piruvāta akumulēšanos Z. mobilis, tika salīdzināti vairāki elpošanas ķēdes mutantie celmi. Uz daļēji inaktivētas piruvātdekarboksilāzes (PDC) fona darbā izdevās iegūt celmu ar pārekspresētu membrānsaistīto D-laktātdehidrogenāzi (LDH), bet neizdevās iegūt celmu ar pārekspresētu NADH dehidrogenāzi. Novērojām, ka elpošanas ķēdes NADH dehidrogenāzes un LDH aktivitātes aerobi augošās Z.mobilis šūnās nesummējas - celmā ar pārekspresētu LDH aktivitāti, bet aktīvu NADH dehidrogenāzi, skābekļa patēriņš notiek ar līdzīgu vai pat mazāku ātrumu nekā celmā Zm6, kurā LDH nav pārekspresēta. Noskaidrojām, ka celmos ar daļēji inaktivētu piruvātdekarboksilāzi elpošana nav limitējošais faktors piruvāta akumulācijā, jo līdzīga piruvāta akumulācija notiek pie dažādiem elpošanas ātrumiem. Lai gan celmiem ar daļēju PDC inaktivāciju ir novērojams 50% kritums tās aktivitātē, lielākā daļa glikozes oglekļa joprojām nonāk etanolā un CO2. SUMMARY To divert catabolism in Z. mobilis from ethanol synthesis to pyruvate synthesis a way to oxidize NADH, that is usually used in ethanol synthesis, needs to be found. Respiration is one of the possible paths of NADH oxidation. Multiple respiratory mutant stains of Z. mobilis where compered to gain an idea of the role that respiratory chain plays in pyruvate accumulation. In this work a strain with overexpressed membrane bound D-lactate dehydrogenase (LDH) on partially inactivated pyruvate decarboxylase (PDC) background was successfully created. An attempt to overexpress NADH dehydrogenase on partially inactivated PDC background was unsuccessful. It was observed that activities of NADH dehydrogenase and LDH in the respiratory chain does not sum-up in aerobically grown Z. mobilis. Furthermore in the strains with overexpressed LDH and active NADH dehydrogenase oxygen uptake reached similar or even lover values than in the ZM6 strain in which LDH was not overexpressed. A conclusion that respiration is not the limiting factor of pyruvate accumulation in Z. mobilis strains with partially inactivated PDC was reached, because similar amounts of accumulated pyruvate were observed in stains with different respiration speeds. Despite the fact that in the stains with partially inactivated PDC a 50% fall in PDC activity can be observed, most of the carbon from glucose is converted into ethanol and CO2.
