C3aR un Cd11b proteīnu ekspresijas noteikšana smadzeņu prefrontālās garozas audos šizofrēnijas genotipa pelēm

Šizofrēnija ir neiroattīstības slimība, kas skar 1% pasaules populācijas pieaugušo vecumā. Šizofrēnijas ārstēšanā izmanto neiroleptiķus, kuri pamatā ietekmē dopamīna un serotonīna līmeņa regulāciju smadzenēs. Diemžēl, šī medikamentu grupa ir efektīva tikai daļai pacientu. Pēdējo gadu pētījumi norāda gan uz GABA sistēmas, gan neiroiekaisuma kaskādes nozīmi šizofrēnijas patoģenēzē (Moslem et al., 2019). Šī darba mērķis ir noskaidrot C3aR un Cd11b ekspresiju kontroles grupas peļu un jaunizveidotā šizofrēnijas peļu genotipa prefrontālās smadzeņu garozas audu lizātos ar Western blot metodi. Rezultāti parāda, ka paraugiem, kuriem bija gēnu knock-out, attiecīgie proteīni netika ekspresēti, norādot, ka jaunie peļu līniju genotipi ir izveidoti. Ņemot vērā jaunākos pētījumus par komplementa sistēmas nozīmi šizofrēnijas attīstībā, radītos modeļdzīvniekus var izmantot tālākā šizofrēnijas slimības raksturošanā, veicot uzvedības, reakciju un organisma bioķīmisko atbildes reakciju izpēti. Schizophrenia is a neurodevelopmental disease that affects 1% of the world's adult population. Neuroleptics, which mainly affect the regulation of dopamine and serotonin levels, are used to treat schizophrenia. Unfortunately, this group of medications is only effective in a subset of patients. Recent studies point out the role of both the GABAergic system and the neuroinflammatory cascade in the pathogenesis of schizophrenia (Moslem et al., 2019). The aim of this study was to determine the C3aR and Cd11b expression in the samples of prefrontal cortex tissue lysates of control groups and the newly developed schizophrenia mouse genotype using Western blot method. The obtained results show that studied proteins were not expressed in the samples of knock-out mice. To conclude, this new knock-out (C3aR and Cd11b) mice strains can used to investigate the complement system in the development of schizophrenia. Further studies using these knock-out mice could involve behavioral tests and to characterization of the biochemical responses in the body.
