Darbā ir apskatīta atomspēku mikroskopijas (AFM) metodes pielietošana cilvēka plaušu biopsijas materiāla izpētei. Sadarbībā ar patologu, izmantojot atomspēku mikroskopu, tika pētīti cilvēka plaušu adenokarcinomas un plakanšūnu karcinomas audu paraugi, kas sagatavoti ar divām metodēm (audu kriosekcionēti griezumi un nospiedumi). Tika apskatīta un salīdzina paraugu morfoloģija, kā arī elastīgo īpašību testi. Veikta šūnu ķīmisko fiksēšanas metožu salīdzināšana, izmantojot adenokarcinomas šūnu A549 kultūru un ASC52telo cilmes šūnu kultūru. No biopsijas iegūto paraugu kvalitātes uzlabošanai un adhēzijas spēku starp zondi un paraugu samazināšanai ar nospiedumu metodi iegūtie paraugi tika ķīmiski fiksēti ar etanolaetiķskābes (3:1) maisījumu. Darbā pētīta AFM mērījumu atkarība no paraugu sagatavošanas metodes un analizēti iegūtie rezultāti. This work reviews the possible applications of atomic force microscopy (AFM) in the study of human lung biopsy material. Within the framework of this course paper, collaborating with a pathologist, human lung adenocarcinoma and squamous cell carcinoma tissue samples, prepared with two methods (cryotomed sections and impression smears) were studied, their morphology was examined and compared using the atomic force microscope. Different cell chemical fixation methods were compared by using A549 cell culture and ASC52telo stem cell culture. To improve the quality of the samples and reduce the probe-sample adhesive forces, the samples obtained by the impression method were chemically fixed with ethanol-acetic acid (3:1) mixture. The dependence of AFM measurements on the sample preparation method is investigated and the results analysed.
