Lunasīna biostabilitāte un citotoksicitāte in vitro testos

Funkcionālā pārtika satur bioloģiski aktīvas vielas ar pierādītu ietekmi uz kādas slimības riska faktoriem. Pārtikas proteīni paši var kalpot kā profilakses līdzekļi, bet papildus tiem enzimātiski sašķeļoties veidojas bioloģiski aktīvi peptīdi. Vispirms sojā, bet pēc tam rudzos, kviešos, miežos un tritikālē atklātais 43 aminoskābju garš peptīds lunasīns spēj preventēt, inhibēt un apstādināt vēža šūnu augšanu, samazināt zema blīvuma lipoproteīnu līmeni asinīs, un tam piemīt pretiekaisuma aktivitāte.
Šī pētījuma mērķis bija izstrādāt lunasīna noteikšanas ELISA metodi un noteikt lunasīna biostabilitāti peļu audos, kā arī pārbaudīt lunasīna citotoksicitāti normālās un cilvēka vēža šūnu līnijās. Iegūtie dati liecina, ka ar ELISA metodi var noteikt lunasīnu koncentrācijā no 20 līdz 120 ng/ml. Pārbaudot lunasīna atgūstamību peļu audu homogenātos, atklājām, ka ar graudu saturošu pārtiku barotām pelēm lunasīns akumulējās nierēs. Lunasīns uzrādīja toksicitāti eksperimentā PA1 un SH-SY5Y šūnu līnijās. Functional foods contain biologically active substances with a preventative effect on a disease risk factors. Food proteins themselves and enzymatically formed biologically active peptides can serve as a prophylactic means. First of all in soy, then in rye, wheat, barley and triticale the 43 amino acid long peptide lunasin with anti-inflammatory action and ability to prevent proliferation and growth of cancer cells was found. Later it was shown that lunasin reduces level of low-density lipoproteins in the blood. This study aimed to develop ELISA method for measurement of lunasin content and to determine biostability of lunasin in mouse tissues as well as to test lunasin cytotoxicity in normal and human cancer cell lines. The obtained data show that with the ELISA assay one can detect lunasin at concentration from 20 to 120 ng / ml. We found that lunasin accumulates in kidney of the grain-based food fed mice. Lunasin showed toxicity in PA1, SH-SY5Y human cancer cell lines used in the experiments.
