Fibroblastu augšanas faktors 2 (FGF-2 jeb bFGF) atrodas gandrīz visos mezodermālās un neiroektodermālās izcelsmes audos un arī to audzējos. Daudzas šūnas izdala FGF-2 īslaicīgi un uzglabā to bioloģiski neaktīvā formā. Vienots mehānisms kā šūnas atbrīvo faktoru nav zināms. FGF-2 un tam radniecīgo faktoru izdalīšana iespējams tiek regulēta dažādi, atkarībā no šūnu veida un attīstības vecuma. Cilmes šūnas, piemēram, to izdala, lai varētu pašatjaunoties.
Bakalaura darba mērķis bija noteikt no cilvēka ādas dermas, adipocītiem un kaulu smadzenēm izdalītu cilmes šūnu FGF-2 sekrēciju, kā arī noteikt vai šūnu stimulācija ar liellopu asins sērumu, melanokortīniem, dibutiril-ciklisko adenozīna monofosfātu (db-cAMF) un putrescīnu ietekmē FGF-2 sekrēciju. Iegūtie rezultāti liecina, ka FGF-2 sekrēcija ir atkarīga no šūnu skaita, db-cAMF inhibē FGF-2 sekrēciju dermas un kaulu smadzeņu cilmes šūnās, bet melanokortīni un putrescīns neizmaina FGF-2 sekrēciju. Tika atklāts, ka no dažādiem audiem iegūtās cilmes šūnas uztur barotnēs samērā konstantu FGF-2 līmeni. Secinājums ir, ka cilmes šūnām piemīt autokrīna/parakrīna FGF-2 sekrēcijas regulācija. Fibroblast growth factor 2 (FGF-2 or bFGF) is found in almost all tissues as well as in tumours. Many cell lineages produce FGF-2 only short time and later save it in biologically inactive form. Unite mechanism how cells release FGF-2 is unknown. Possibly, it depends on cell cycle, aging and stage of development. Stem cells release FGF-2 for self-renewal.
Goal of the bachelor thesis was to study human dermal, adipocyte and bone marrow stem cell capacity to secrete FGF-2, as well as to determine, do fetal bovine serum (FBS), melanocortins, dibutyryl-cyclic adenosine monophosphate (db-cAMP) and putrescine influence the cell FGF-2 release. Obtained results show that FGF-2 secretion depends on cell number, db-cAMP inhibits FGF-2 release from dermal and bone marrow cells, but melanocortins and putrescine do not influence the release of FGF-2 in the cell media. We observed quite constant and equal FGF-2 secretion capacity of the human dermal, adipocyte and bone marrow stem cells. We suggest autocrine/paracrine regulation of FGF-2 secretion.
