Adenozīna A3 receptors pieder ar G – proteīnu saistīto receptoru grupai, kurai ir liela nozīme fizioloģiskajās funkcijās un ir daudzu farmakoloģisko pētījumu mērķis.
Šis ir pirmais pētījums, kurā salīdzina adenozīna A3 receptora wild type un cilvēku populācijā sastopamā receptora mutanta aktivitāti. Darbā ir veikta pētāmā receptora kodējošās sekvences maiņa un sajūgšana ar zaļi fluorescējošo proteīnu. Darbā tika noskaidrota polimorfisma ietekme uz receptora aktivitāti. Tāpat noskaidrots, ka GFP nav piemērots adenozīna A3 lokalizācijas pētījumiem.
Darba mērķis ir adenozīna A3 receptora cilvēka populācijā sastopamā nesinonīmā SNP rs35511654 ietekmes uz receptora aktivitāti un receptora lokalizāciju noteikšana.
Darbs veikts no 2012. gada oktobra līdz 2013. gada maijam Latvijas Biomedicīnas pētījumu un studiju centrā.Atslēgas vārdi: Adora3, PCR virzītā mutaģenēze, I248L, rs35511654, GFP. A3 adenosine receptor belongs to the G - protein coupled receptor group, which plays an important role in many physiological functions and is the target in pharmacological researches.
This is the first study that compares the adenosine A3 receptor wild type human and wild-mutant receptor activity. Receptor coding sequence change and connection to green fluorescent protein has been carried out in this study. This work studied and compared the effect of polymorphism on the receptor activity and that GFP is not suitable for A3 adenosine receptor localization studies.
The aim of this research is to determine the effect of common non-synonymous SNP rs35511654 on activity and receptor localization.
