Darbā risināta selekcionāru novērota problēma: pēc pārsējas dubultoto haploīdu (DH) pēcnācējos ir dažādība, bet DH līnijām jābūt 100% homozigotiskām. Dažādība, iespējams, saistīta ar to, ka no putekšņu maciņiem iegūtos embriju kopas veido daudzi kopā salipuši putekšņi, kam katram atsevišķi novērojama embrioģenēze. Embrijus kultivēšanas laikā dalot jaunie augi-reģeneranti veidotos no atsevišķiem embrijiem.
Ziemas kviešu putekšņmaciņu kultūrās in vitro iegūti mikrosporu veidoti embriji, kas tika dalīti. Izaudzētie haploīdi augi, hromosomu komplekta dubultošanas nolūkā, apstrādāti ar kolhicīnu. Iegūtās fertilās DH līnijas nodotas selekcionāriem izvērtēšanai lauka apstākļos.
Ar PCR pierādīts, ka dažas embriju kopas ir ģenētiski heterogēnas, un, acīmredzot, veidojušās no atšķirīgām mikrosporām – embriju kopu dalīšana novērš dažādību DH līnijās. The work solves problem observed by breeders: diversity of progeny of doubled haploids (DH), but the DH lines must be 100% homozygous. Diversity is probably due to the fact that the pollen from the anthers of the resulting embryo is composed of numerous sets of pollen stuck together, each individual observed in embryogenesis.
From winter wheat hybrids anther cultures in vitro were obtained microspores formed embryos, which were divided. From the divided embryos were grown separate haploid plants-regenerants, which, for duplication of chromosome set, were treated with colchicine. Obtained fertile DH lines planted in soil for evaluation under field conditions.
PCR demonstrated that some clusters of embryos are genetically heterogeneous, and, apparently, formed from different microspores - dividing of embryonic clusters eliminates the diversity of DH lines.
