Hloroplastu telpiskās struktūras un funkcionālās aktivitātes atšķirības atkarībā no to novietojuma šūnā

Bakalaura darba mērķis bija Raksturot hloroplastu telpisko struktūru un hlorofila fluorescences izmaiņas novecošanās laikā dažāda vecuma tabakas lapās.
Par dažāda vecuma lapām izvēlējās mazas lapas (7 mm), jaunas (3 cm), pieaugušas un novecojušas (15 cm) lapas. No lapām tika pagatavoti preparāti lāzera skenējošajai konfonkālajai un skenējošajai elektronu mikroskopijai. Preparātos novēroja šūnas un to organellas – hloroplastus un kodolus, to savstarpējo mijiedarbību. Hloroplastiem raksturoja to biezumu, telpisko novietojumu, formu, membrānu izaugumus, fluorescences atšķirības un novietojumu šūnā.
No iegūtajiem rezultātiem var secināt, ka šūnu organellas savstarpēji veido dažādus kompleksus, piemēram, kodoli ar hloroplastiem. Hloroplastu biezums un diametrs mainās laikā dažāda vecuma lapās. Jaunām lapām raksturīgi biezi lēcveida hloroplasti ar mazu diametru un retiem izaugumiem uz membrānas, bet vecās lapās to biezums samazinās un diametrs palielinās.
Pieaugušām lapām plātnes apikā The aim of the Bachelor was to characterize and study the 3D structure of chloroplasts in tobacco leafs of different ages, and to identify changes in chloroplast ultra structure and position in the cells. And to identify chlorophyll fluorescence in chloroplasts, which are placed in the periphery and to the nucleus of the cells in tobacco leaves of different ages.
For different ages were choosen small (7 mm), young (3 cm), adult and old leafs (15cm). From these leafs was prepared preparations for laser scanning confocal and scanning electron microscopy. Cells and their organelles were observed– the interaction of chloroplast and nuclei. In chloroplasts were described their thicknesses, spatial location, shape, membrane knobs and difference of fluorescence and position in the cell.
From the results it is concluded that the cell organelles forms variety of complex between each other, such as the nuclei and chloroplast complexes. The shape and thickness of chloroplasts varies at differen
