Haloperidola un mildronāta efekti uz biomarķieru ekspresiju hipokampā žurkām

Atmiņa ir process, kura pamatā ir sinaptiskās plasticitātes procesi dažādos smadzeņu apgabalos. Atmiņas pasliktināšanās iemesli var būt dažādi – vecums, slimības, medikamenti u.c. Antipsihotiskajam medikamentam haloperidolam piemīt spēja negatīvi ietekmēt atmiņu. Iespējams, ka mildronāts šos bojājumus varētu novērst.
Šajā darbā ar Western blota metodi tika pētīta haloperidola (1 mg/kg) un mildronāta (50 un 100 mg/kg) ietekme uz biomarķieru: smadzeņu atvasinātā neirotrofiskā faktora (BDNF), acetilholīnesterāzes (AChE) un glutamāta dekarboksilāzes 67 (GAD67) ekspresiju žurku hipokampā pēc 3 nedēļu vielu ievadīšanas.
Iegūtie rezultāti uzrāda BDNF un AChE ekspresijas samazināšanos haloperidola ietekmē. Mildronāta premedicēšana atgrieza BDNF un AChE kontroles līmeņos. Ņemot vērā iegūtos rezultātus var teikt, ka mildronātam ir potenciāls aizsargāt no haloperidola izraisītas amnēzijas žurkām.

Reasons for memory impairment can be different – aging, diseases, drugs and other. An antipsychotic drug – haloperidol – shows ability to effect memory negatively. Might be that mildronate could reverse these lesions.
In this study, using the Western blot method was examined the effect haloperidol (1 mg/kg) and mildronate (50 and 100 mg/kg) on expression of biomarkers: brain derivered neurotrophic factor (BDNF), acetylcholinesterase (AChE) and glutamic acid decarboxylase 67 (GAD67) in rat hippocampus, after 3 weeks of premedication.
The Western blot data show that haloperidol have decreased levels of BDNF and AChE. Premedication of mildronate returned BDNF and AChE to the levels of control group. Considering obtained results, we may say that mildronate has the potential to preserve rat’s memory from haloperidol caused amnesia.
