Rezistento baktēriju celmu straujā izplatība rada nopietnu draudu cilvēku veselībai visā pasaulē. Aminoacil-tRNS sintetāzes (aaRS) ir daudzsološs, bet nepietiekami izmantots, terapeitiskais mērķis jaunu antibakteriālu līdzekļu izveidei. Lai atrastu jaunus aaRS inhibitorus, darbā veikts 1000 fragmentu tipa savienojumu bibliotēkas skrīnings uz saistību pie izoleicil-tRNS sintetāzes (IleRS), izmantojot KMR spektroskopiju. Identificētajiem IleRS trāpījumiem tālāk pārbaudīta saistība ar divām citām aaRS, un veikta trāpījumu savstarpējās konkurences pārbaude. Darbā atrastie plaša spektra inhibitori ir augstvērtīgi trāpījumi, kurus tālāk plānots attīstīt par spēcīgiem un patentspējīgiem aaRS inhibitoriem.

The increasing spread of antibiotic-resistant bacterial strains is a major public health threat worldwide. Aminoacyl-tRNA synthetases (aaRS) are a very promising, but unexploited group of antibacterial drug targets. In order to discover novel aaRS inhibitors a library of 1000 fragment-type compounds was screened using NMR spectroscopy against binding to isoleucyl-tRNA synthetase (IleRS). The identified IleRS hits were further evaluated against binding to two other aaRS, as well as tested for competition between each other. The identified multi-target binders are high-quality hits that will be optimized into potent and patentable lead compounds.
