IS10 ir viens no aktīvākajiem transpozoniem, kas nodrošina adapatīvo mutaģenēzi baktērijām, aktivizējot neekspresējošus gēnus. Darba mērķis bija noskaidrot hloramfenikola selektīvā spiediena ietekmi uz IS10 elementa spēju efektīvāk aktivizēt bezpromotera acetiltransferāzes (cat) gēnu pAdCat plazmīdā Escherichia coli kultūras stacionārā augšanas fāzē. Baktēriju celmi tika transformēti ar pAdCat plazmīdu, kas kalpo par vektoru-lamatu IS10 elementu izolēšanai pēc pozitīvās selekcijas principa. Šūnām, atrodoties trīs dienas stacionārajā fāzē, novēroja IS10 transpozīciju un bezpromotera hloramfenikola acetiltransferāzes (cat) gēna aktivizēšanu. Gēna aktivizēšanu nodrošināja gan IS10R, gan IS10H. Sekvenēšanas un reālā laika PCR rezultāti parādīja, ka hloramfenikola selektīvā spiediena ietekmē IS10 transpozēja tuvāk cat apgrieztā orientācija, piešķirot šūnām izteiktu hloramfenikola rezistences fenotipu ar augstu cat ekspresiju. IS10 is one of the most active transposons in bacteria that provide adaptive mutagenesis by activating silenced genes. The aim of the work was to find out chloramphenicol selective pressure effect on the acetyltransferase gene activation in pAdCat plasmid of the Escerichia coli culture in stationary growth phase. Bacterial strains were transformed with a plasmid pAdCat, which serves as a vector-trap for IS10 element isolation by positive selection principle. In transformants that were kept in stationary phase for three days observed IS10 transposition and chloramphenicol acetyltransferase (cat) gene activation. Gene activation provided by IS10R and IS10H. Sequencing and real-time PCR results showed that by chloramphenicol selective pressure IS10 transpose closer to the cat in the reverse orientation, giving cells strong chloramphenicol resistance phenotype with high cat expression.
