Šūnu līniju iegūšana no krūts vēža punkcijas biopsijas paraugiem

Lai raksturotu ļaunadbīgu krūts audzēju veidošo atšķirīgo šūnu populāciju un noteiktu rezistences pret pielietoto terapiju iemeslus, nepieciešama metode, ar kuru būtu iespējams izdalīt maksimāli daudz šūnu no pieejamā krūts vēža bioloģiskā materiāla. Darba mērķis bija iegūt primārās krūts vēža šūnas no punkcijas biopsijas parauga un veikt to pirmējo raksturošanu.
No trīs biopsiju materiāliem tika izdalītas primārās krūts vēža šūnas un izsētas atšķirīgā blīvumā. Tās tika pavairotas un veikta pirmējā raksturošana, izmantojot gēnu ekspresijas analīzi un imūncitoķīmijas metodi.
Darbs izstrādāts RSU Onkoloģijas institūta Molekulārās ģenētikas laboratorijā un PSKUS Šūnu transplantācijas centrā no 2014. gada septembra līdz 2015. gada maijam. Breast cancer is made up of diverse cell population and some of them may be resistant to the therapy used. Therefore it is essential to develop a method for obtaining utmost number of cells from breast cancer tissue samples. The aim of the work was to obtain primary cells of breast cancer core biopsies and make basic characterisation of them.
Primary breast cancer cells were isolated from three core biopsy matherials and seeded in different densities. Cells were cultivated and primary characterisation was done, using gene expression analysis and imunocytochemistry.
This work was performed in RSU Institute of Oncology Laboratory of Molecular Genetics and PSKUS Cell Transplantation Center and RSU Laboratory of the Molecular Genetics from September 2014 till May 2015.
