Clostridium ģints baktēriju noteikšana dzeramajā ūdenī izmantojot Fluorescento in situ hibridizāciju

Bakalaura darba mērķis bija ātri un precīzi identificēt dzeramajā ūdenī Clostridium ģints baktērijas izmantojot Fluorescento in situ hibridizāciju. Līdz ar to tika izmērīts un salīdzināts fluorescences intensitātes līmenis dažādām Clostridium sugām, kā arī citiem mikroorganismiem izmantojot Clostridium spp. specifiskas fluorescentās zondes. 92% no Clostridium acetobutylicum izmērītam šūnām, relatīvās gaismas vienības (RLU) pārsniedza 500 vienību robežu, 99% no Clostridium beijernickii un 96% no Clostridium tetahomorphum, salīdzinājumā ar Sphingomonas paucimobilis, kur tikai 2% pārsniedza 300 RLU. Veiktais variācijas analīzes tests (ANOVA) liecināja, ka starp Clostridium ģints baktērijām un Sphingomonas paucimobilis pastāv būtiska fluorescences atšķirība (p<0.05). Papildus tam tika sagatavoti un analizēti mākslīgi piesārņoti paraugi, kuros izdevās precīzi noteikt pievienoto klostrīdiju koncentrāciju. Balstoties uz iepriekš iegūto informāciju par šūnu fluorescenci, tika analizēti 12 dažādas izcelsmes Latvijas dzeramā un virszemes ūdens paraugi. Mikroskopēšanai tika izmantota metodika, kas nodrošina iespēju aplūkot 1/5 daļu mikroskopa parauga laukuma un tādējādi samazināt noteikšanas robežas līdz 5 šūnām analizējamā parauga tilpuma vienībā. Vides paraugu analīžu rezultāti parādīja, ka Clostridium ģints pārstāvji nav sastopami. Kopumā metodei vēl ir nepieciešami uzlabojumi, lai galvenokārt samazinātu fona fluorescences intensitāti un palielinātu šūnu fluorescences intensitāti, īpaši, ja tiek analizēti ļoti piesārņoti paraugi. The aim of this study was to identify Clostridium species in the drinking water, using Fluorescent in situ hybridization. Using Clostridium spp. specific fluorescent probes, the fluorescence intensity, between various Clostridium species and other microorganisms, was compared. As a result, 92% of Clostridium acetobutylicum cells exceeded the limit of 500 relative light units (RLU), as well as 99% of Clostridium beijernickii and 96% of Clostridium tetahomorphum, compared to Sphingomonas paucimobilis, where only 2% of cells were higher than 300 RLU. Hence, there is a significant difference between the fluorescence intensities of Clostridium species and Sphingomonas paucimobilis cells (p<0.05, ANOVA test). Additionally, artificially contaminated samples were prepared and analysed. It was possible to accurately identify the added Clostridium concentration. On the basis of the information obtained before, 2 natural drinking and surface water samples from various places in Latvia were analysed. The chosen microscopy method provided the ability to overview 1/5 of total membarne area, thus, at the same time reduced the detection limit up to 5 cells per volume unit of the tested sample. The analysis of environmental samples showed, that the Clostridium genus cells were not detected. However, the methodology is still limited by a lack of fluorescence intensity of positive cells. The improvements are required to reduce the intensity of the fluorescence background, especially if heavily contaminated samples are analysed.
