Kultivēšanas apstākļu ietekme uz ekstracelulāro fruktānu veidošanos Gluconobacter nephelii A14 kultūrā

Mikroorganismu sintezētie ekstracellulārie fruktāni ir salīdzinoši nesen kļuvuši komerciāli pieejami, un tiem ir plašs pielietojums biotehnoloģijā. Šobrīd fruktānu pielietojumu ierobežo to iegūšanas augstajās izmaksas. Tādēļ aktuāli ir pētījumi saistībā ar jaunu fruktānu sintezējošu mikroorganismu kultūru meklējumiem un jau esošo eksopolisaharīdu ieguves metožu pilnveidošanu.
Darba gaitā noskaidrots, ka būtiskākie faktori, kas ietekmē šūnu augšanu un fruktānu veidošanos Gluconobacter nephelii A14 kultivācijā ir saharozes un rauga ekstrakta sākuma koncentrācijas un aerācija. Apstiprināts, ka saharozes un rauga ekstrakta koncentrāciju efekti ir atkarīgi no aerācijas intensitātes. Izveidoti daudzfaktoru lineārās regresijas modeļi, kuri var prognozēt G. nephelii produktivitāti pie atšķirīgām kultivācijas apstākļu gradācijām.
Bakalaura darbs izstrādāts Latvijas Universitātes Mikrobioloģijas un biotehnoloģijas institūtā Tehniskās mikrobioloģijas un Pārtikas biotehnoloģijas laboratorijā laika posmā no 2014. gada septembra līdz 2015. gada jūnijam.
Kursa darba apjoms ir 51 lpp. Tas satur 14 tabulas un 15 attēlus. Microbial synthesis of extracellular fructans only recently has become commercially available, and they have a broad range of applications in biotechnology. Currently the use of fructans are limited due to high acquisition costs. It is therefore an actual task of searching new fructan-producing microorganisms and improve the existing exopolysaccharides processing methods.
During this study it was shown that the sucrose and yeast extract concentration in the culture medium and agitation intensity are the most important factors affecting cell growth and fructan production by Gluconobacter nephelii A14. A significant interaction between factors was confirmed where the effect of sucrose and yeast extract concentration depends on the agitation intensity. Multivariate linear regression models have been developed that can predict the productivity of G. nephelii A14 at different gradations of cultivation conditions.
The bachelor work was developed in the Laboratory of Industrial Microbiology and Food Biotechnology of the Institute of Microbiology and Biotechnology, University of Latvia, from September 2014 until June 2015.
