Ģenētisko faktoru asociācija ar Helicobacter pylori seroloģisko prevalenci Latvijas populācijā

Baktērija Helicobacter pylori (HP) izraisa hronisku gastrītu, peptisko čūlu, kuņģa adenokarcinomu, MALT limfomu, un tās radītā infekcija ir visplašāk izplatītā cilvēku infekcija. Populācijas pētījumā atklāts, ka HP prevalence Latvijā ir gandrīz 80%, kas ir augsta.
Pamatojoties uz Nīderlandes GWAS pētījumu rezultātiem, tika izvēlēti 5 gēni un to polimorfismi- PCLO (rs147900026), TLR10 (rs12233670), FCGR3A (rs147174426), TLE4 (rs138776142), ADRA1D (rs6107461)- lai noskaidrotu, vai šie polimorfismi asociējas ar HP seroprevalenci un kuņģa gļotādas atrofiju Latvijas populācijā.
Rezultāti: PCLO gēna rs147900026 G alēle samazina HP infekcijas risku, FCGR3A gēna rs147174426 T alēle tiek būtiski asociēta ar kuņģa gļotādas atrofijas attīstības risku HP- indivīdiem un TLR10 gēna rs12233670 protektīvā T alēle vairāk raksturīga paraugkopai ar pepsinogēna attiecību PgI/PgII>2.
Secinājumi: Starp pētāmajiem polimorfismiem, rs147900026, rs147174426, rs12233670 var būt nozīme HP infekcijas un smagas kuņģa gļotādas atrofijas patoģenēzē. Helicobacter pylori (HP) is a common gastric pathogen that causes gastritis, peptic ulcer disease, gastric adenocarcinoma, and low-grade gastric lymphoma and it represents the most widespread infection in the world. The prevalence of HP in Latvia is almost 80%, which is high.
Based on the GWAS in Netherlands, we chose 5 genes and their polymorphisms- PCLO (rs147900026), TLR10 (rs12233670), FCGR3A (rs147174426), TLE4 (rs138776142), ADRA1D (rs6107461)- to determine, if these polymorphisms are associated with HP seroprevalence and gastric mucosal atrophy in Latvian population.
Results: PCLO gene rs147900026 G allele decrease HP infection risk, FCGR3A gene rs147174426 T allele is significantly associated with gastric mucosal atrophy development risk in HP- individuals and TLR10 gene rs12233670 protective T allele is more common in sample group with pepsinogen ratio PgI/PgII>2.
Conclusions: rs147900026, rs147174426, rs12233670 SNPs may play a role in the pathogenesis of HP infection and severe gastric mucosal atrophy.
