Alcheimera slimības modelēšana žurkām ar streptozotocīna intracerebroventrikulāro ievadīšanu

Šī darba mērķis bija izveidot žurku STZ icv AD-tipa modeli un pārbaudīt GABAA agonista muscimola mazo devu (0,01 un 0,05 mg/kg) ietekmi uz iemācīšanos un atmiņu ūdens labirinta testā.
Rezultāti rāda, ka:
a) Abās MUSC-aCSF grupās izbēgšanas latence treniņdienu laikā samazinājās līdzīgi kontrolei; efekti saglabājās arī gala testā;
b) Abās MUSC-STZ grupās izbēgšanas latence treniņdienās būtiski samazinājās pret Sal-STZ (p<0,0001), un gala testā būtiski pieauga platformas zonas šķērsošana un laiks platformas kvadrantā pret Sal-STZ (p<0,0001).
Šie dati liecina, ka STZ icv ievade spēj ietekmēt AD-tipa agrīnos kognitīvos traucējumus un muscimols spēja tos reversēt caur GABA aktivēšanos. GABAerģisko procesu regulācija sniedz jaunas iespējas anti-demences terapiju izstrādē. The aim of the present study was to create rat icv STZ AD-type model and test the effects of GABAA agonist muscimol (0.01 and 0.05 mg/kg) on learning and memory in the water maze task.
The results show that:
a) Escape latencies in both MUSC-aCSF groups notably decreased in training days similar to control and effects were maintained in the probe trial;
b) Escape latencies in both MUSC-STZ groups notably decreased in training days vs. Sal-STZ (p<0.0001); platform zone crosses (p<0.0001), time in platform quadrant (p<0.0001) notably increased vs. Sal-STZ.
The data show that icv STZ can enhance AD-type cognitive dysfunction and muscimol via activation of GABA was able to reverse it. Regulation of GABAergic processes is an intriguing step in the search of new anti-AD therapies.
