Darba mērķis bija jaunu sekundāro metabolītu izolēšana no sūkļa Smenospongia aurea un to ķīmiskās struktūras noteikšana. Tomēr, uzsākot darbu, nolēmām izdalīt oriģinālus, neribosomālus hibrīdu peptīdu/poliketīdu savienojumus – smenamīdu A, smenamīdu B, smenotiazolu A un smenotiazolu B –, kas jau iepriekš izdalīti no S. aurea sūkļa un kam noteikta augsta citotoksiska aktivitāte. Interesējošos savienojumus izdalījām no lipofīla sūkļa S. aurea ekstrakta, izmantojot augsti efektīvo šķidrumu hromatogrāfijas (AEŠH) metodi, to ķīmisko struktūru noteicām ar augstas izšķirtspējas masspektrometrijas (ESI/MS), 1H kodolu magnētiskās rezonanses (KMR) un 13C KMR metodēm, un iegūtos datus salīdzinājām ar literatūras datiem.
Rezultātā izdevās izdalīt individuālus savienojumus – smenamīdu A, smenamīdu B, smenotiazolu A un smenotiazolu B – ļoti mazos daudzumus, ko var skaidrot ar bromēto alkaloīdu daudzumu S. aurea sūklī, kas apgrūtina sekundāro metabolītu hromatogrāfisku atdalīšanu. The present study was dedicated to the isolation of novel secondary metabolites from the Carribean sponge Smenospongia aurea and structure elucidation of compunds. However, during the process we decided to isolate original hybrid peptide/polyketide secondary metabolites of S. aurea smenamide A, smenamide B, smenothiazole A, smenothiazole B, which were already described in literature as potent cytotoxic compounds. High-performance liquid chromatography (HPLC) method was implemented to isolate ompounds of interest from the lipophilic extract of S. aurea. High-resolution electrospray ionization tandem mass spectrometry (ESI/MS) method, 1H nuclear magnetic resonance (NMR) and 13C NMR methodes were implemented to elucidate isolated compounds. Data were compared with those described in literature.
As a result we isolated individual compounds smenamide A, smenamide B, smenothizaloe A and smenothiazole B in a very minute amounts, partly because they were very difficult to separate chromatographically from brominated alkaloids, which are present in S. aurea.
