Zaru apstrādes un fitohormonu ietekme uz smaržīgā ķirša Prunus mahaleb un skābā ķirša Prunus cerasus ziemojošo pumpuru attsītību in vitro

Ķirši ir plaši izmantoti kultūraugi. Tirgū tie ir pieprasīti, tāpēc ir svarīgi noskaidrot, kā pēc iespējas efektīvāk pavairot attiecīgās ķiršu šķirnes stādus. Pavairojot ķiršus ar potēšanas metodi, jāņem vērā, ka potcelms var ietekmēt uzpotētās šķirnes īpašības, piemēram, ziemcietību, koka augumu. Potcelms, kurš palielina ziemcietību un samazina augumu, ir vērtīgs, audzējot ķiršus komerciālos nolūkos. Šī darba mērķis bija noskaidrot smaržīgā ķirša un skābo ķiršu šķirnes ‘Latvijas zemais’ zaru apstrādes pirms ievadīšanas audu kultūrā un barotnei pievienoto fitohormonu ietekmi uz ziemojošo pumpuru attīstību in vitro. Lai šo mērķi sasniegtu, izvirzīja sekojošos uzdevumus: sterilizēt un ievadīt smaržīgā ķirša pumpurus Murashige & Skoog (MS) barotnēs, plaucēt citus smaržīgā ķirša un skābā ķirša zarus pirms pumpuru ievadīšanas MS barotnēs fitohormonu benzilaminopurība (BAP) (0,5 mg L-1), naftiletiķskābes (NES) (0,5 mg L-1) šķīdumos, dejonizētā ūdenī vai saharozes šķīdumā (30 g L-1), iegūtos dzinumus ievadīt ar fitohormoniem BAP (0,5 mg L-1) vai BAP (0,5 mg L-1) ar indolsviestskābi (ISS) (0,1 mg L-1) bagātinātās barotnēs, no kurām barotnē ar BAP un ISS notika vislabākā dzinumu attīstība. Cherries are broadly grown as cultivated plants. Because of high demand for cherries on the market it is necessary to find out how to effectively propagate cherry cultivars. By propagating cherries with grafting, it must be taken into account that the rootstock can affect the traits of the grafted cultivar, for example, winter hardiness, growth. A rootstock which increases winter hardiness and reduces growth is valuable when cherries are grown commercially. The aim of this work was to determinate the influence of twig preparation and phytohormons on St. Lucie cherry’s and sour cherry cultivar’s ‘Latvijas Zemais’ development of overwintering buds in vitro. To achieve this aim, some buds of St. Lucy cherry were sterilized and introduced in Murashige & Skoog (MS) media, before other buds of St. Lucie cherry and the buds sour cherry were introduced in (MS) media, buds were sprouted in phytohormon benzylaminopurine (BA) (0.5 mg L-1), naphthyl acetic acid (NAA) (0.5 mg L-1) solutions, deionised water or sucrose (30 g L-1) solution. The obtained shoots were introduced in a MS medium supplemented with phytohormons (BA (0.5 mg L-1) or BA (0.5 mg L-1) with indolbutyric acid (IBA) (0.1 mg L-1)). Best growth of shoots in vitro was achieved on medium with BA and IBA.
