Antioksidanti ir plaši izplatīti augu valsts pārtikas produktos un pēdējā desmitgadē tiem ir pievērsta pastiprināta uzmanība. Pateicoties to veselību uzlabojošajām īpašībām, ir radusies palielināta interese par to noteikšanas veidiem dažāda veida augos un augu izcelsmes produktos. Līdz ar augsti efektīvās šķidrumu hromatogrāfijas pieejamību apvienojumā ar augstas izšķirtspējas masspektrometriju, ir pavērti jauni apvāršņi analītisko metožu izstrādei sarežģītās paraugu matricās. Darbā dots ieskats par antioksidantu un polifenolu īpašībām, to izdalīšanu no pārtikas produktiem, paraugu sagatavošanas un noteikšanas metodēm, izmantojot augsti efektīvās šķidruma hromatogrāfijas metodes. Kā pētījumu objekts ir izvirzīti antioksidanti, kas ir sastopami šokolādē un citos kakao produktos. Darbā ir izstrādāta un pielietota antioksidantu analīzes metode izmantojot hibrīda-Orbitrap augstas izšķirstspējas masspektrometru. Izstrādātas paraugu sagatavošanas metodes, kuras ir ātras un efektīvas. Metode validēta, lai pārliecinātos par metodes precizitāti, pareizību un kopējo uzticamību. Noteikts antioksidantu saturs 26 šokolādes paraugos, kuros kopējais 8 izvēlētu antioksidantu saturs ir robežās no 330 – 2631 mg·kg-1 parauga. Antioxidants are wildly distributed in plant products and in recent decades an attention has been raised due to their beneficial properties to human health. An interest has been given in determination of polyphenolics in different types of plants and plant products. Consequently, availability of UPLC systems and columns with sorbent sizes smaller than 2 μm and the modern high resolution mass spectrometry tools indicate new horizons to the development of new methods for the analysis of complex sample matrices. The study gives an overview of antioxidant and polyphenol charateristic properties, distribution in food products, sample preparation and detection methods, using HPLC. As the main subjects of research antioxidants which are found in chocolate and other cocoa products have been evaluated. The developed method is based on rapid sample preparation and UPLC methods in tandem with hybrid-Orbitrap HRMS technology. Method has been validated to assure its precision, accuracy and reliability. 26 chocolate samples were analysed and concentrations of selected 8 antioxidants were determined. Total concentrations of selected antioxidants varied from 330 to 2631 mg·kg-1.
