Clostridium perfringens identificēšana un uzskaitīšana dzeramajā ūdenī ar fluorescentās in situ hibridizācijas metodi

Sporu-veidojošā C. perfringens ir nozīmīgs organisms ūdens vidē, ko izmanto kā indikatororganismu dzeramā ūdens kvalitātes noteikšanai. Turklāt šī baktērija un tās sporas ir noderīgas, lai brīdinātu par citu stresa rezistento patogēnu iespējamo klātbūtni dzeramajā ūdenī. Atbilstoši Ministru kabineta noteikumiem Nr. 235, Clostridium perfringens noteikšanai un uzskaitei jāizmanto membrānfiltrācija un tālāka mikroorganismu kultivēšana. Tomēr izpētīts, ka kultivēšanas metodes nespēj identificēt ūdenī esošus nekultivējamus bet dzīvotspējīgus mikroorganismus (VBNC). Fluorescentā in situ hibridizācija (FISH), nodrošina baktēriju identificēšanu to dabiskā vidē, un var būt izmantojama, ka alternatīvā metode. Taču standarta FISH protokols nav piemērots baktēriju sporu identificēšanai. Tas ir skaidrojams ar to, ka C. perfringens sporas ir ļoti izturīgas pret dažādiem nelabvēlīgiem apstākļiem, to starp dezinfekciju, UV apstarošanu, ķīmisko vielu apstrādi, kā arī pret dažādām caurlaidību pastiprinošām procedūrām. Līdz ar to C. perfringens sporas var ilgstoši saglabāties ūdens apgādes sistēmā neidentificētas. Šajā pētījumā izstrādātais permeabilizācijas protokols, kur sporas jāinkubē 30 minūtes (70˚C) 5% nātrija dodecilsulfāta un 50 mM detiotreitola maisījumā, ar tālāko 15 minūtes (37˚C) 10 mg/ ml vai 20 mg/ ml lizocīma apstrādi, palielināja C. perfringens sporu fluorescences intensitāti deviņas reizes, kur sporu fluorescence sasniedza 1800 RLU. Apvienojot izstrādāto permeabilizācijas protokolu ar Fluorescento in situ hibridizāciju, arī dabiskajos paraugos bija iespējams identificēt Clostridium perfringens veģetatīvas šūnas un sporas ar fluorescences intensitāti virs 1000 RLU. Spore-forming Clostridium perfringens has an important ecological role in aquatic environments and is used as an indicator of drinking water quality. According to ISO 14189:2013, the enumeration of vegetative cells and spores of C. perfringens is performed by the membrane filtration method, which is based on cultivation. This method is time consuming and can not enumerate nonculturable cells. An alternative method is fluorescence in situ hybridization (FISH) which allows in situ detection of target cells in their native environments without cultivation. Nevertheless, typical FISH protocol needs to be improved and modified if bacterial spores are to be detected. All Clostridia spores are extremely resistant to harsh conditions, including disinfection, chemicals and UV light, which means they can persist through the disinfection process and last for long periods in water supplies. At the same time fluorescent probe binging is limited due to reduced accessibility. Since no universal or standard spore permeabilization procedure was available in the literature, the aim of this study was to develop an effective protocol to permeabilize C. perfringens spores in order to apply the FISH method. The most effective combination, 5% Sodium dodecyl sulfate and 50 mM Ditiotheitol mixture, with combination of 10 mg/ ml Lysozyme treatment, allowed to obtain increased spore fluorescence signals and demonstrated a technology for the detection of Clostridium cells and spores in less than 6 hours. However, in comparison with alternative method (qPCR), FISH method demonstrated insufficient sensitivity, that prevented exact quantities determination of C. perfringens in biofilm.
