Mitohondriju galvenā funkcija ir enerģijas ražošana šūnā. Traucējumi mitohondriju elpošanas ķēdē var izraisīt plaša spektra patoloģijas. Patogēno mtDNS mutāciju ietekmi uz šūnu fenotipu iespējams pētīt citoplazmatisko hibrīdu šūnu līnijās. Darba mērķis ir analizēt citoplazmatisko hibrīdu šūnu modeļus un noteikt to piemērotību mitohondriālo slimību raksturošanai un diagnostikai. Tika veikta kontroles grupas un pacientu citoplazmatisko hibrīdu šūnu līniju mtDNS fragmentu sekvenēšana, mtDNS kopiju skaita, ATF koncentrācijas un elpošanas ķēdes kompleksu aktivitāšu noteikšana un salīdzināšana, kā arī tika raksturota izveidoto šūnu līniju stabilitāte. Lai apstiprinātu atbilstību donoru trombocītu mtDNS, tika veikta mtDNS fragmentu sekvenēšana 28 kontroles grupas šūnu līnijām un 12 pacientu šūnu līnijām. Tika noteikts mtDNS kopiju skaits 38 kontroles un 7 pacientu šūnu līnijām. ATF koncentrācija tika analizēta 7 kontroles un 8 pacientu līnijās. Elpošanas ķēdes kompleksu aktivitāte tika pārbaudīta divās šūnu līnijās un salīdzināta ar laboratorijā iepriekš veiktajiem mērījumiem. Citoplazmatisko hibrīdu šūnu modeļu stabilitātes raksturošanai, iepriekš minētie mērījumi tika veikti un salīdzināti starp vienas līnijas dažādu pasāžu šūnām. The main function of mitochondria is to produce energy for the cell. Interference in mitochondrial respiratory chain can cause a broad spectrum of pathology. The influence of pathogenic mtDNA mutations on the cell phenotype can be studied using cytoplasmic hybrid cell lines. The aim of this study was to analyze cytoplasmic hybrid cell models and determine their suitability for mitochondrial disease characterization and diagnostics. Control group and patient cytoplasmic hybrid cell line mtDNA fragments were sequenced and the number of mtDNA copies, ATP concentration and respiratory chain complex activity was determined and compared. Additionally, stability of constructed cell lines was assessed. Twenty eight control group and 12 patient cell line mtDNA fragments were sequenced in order to confirm their match to the platelet mtDNA. Number of mtDNA copies was determined in 38 control and 7 patient cell lines. ATP concentration was analyzed in 7 control and 8 patient cell lines. Respiratory chain complex activity was assessed in two cell lines and compared with previously obtained measurements in the same laboratory. To characterized cytoplasmic hybrid cell model stability, previously described parameters were determined and compared within one cell line different passages.
