Zymomonas mobilis elpošanas ķēdes mutantu skābekļa un temperatūras stresa tolerance

Zymomonas mobilis Zm6 celms un astoņi tā elpošanas ķēdes mutantu celmi tika audzēti četros dažādos apstākļos: 30oC aerobi (kratītāja kolbās), 30oC mikroaerobos (bez kratīšanas), 42oC aerobi un 42oC mikroaerobi, lai salīdzinātu oksidatīvā un termālā stresa ietekmi uz katru no šiem celmiem. Tika noteikta celmu augšana, etanola produkcija un skābekļa patēriņš. Citohroma c peroksidāzes pārekspresija uzlaboja augšanu 30oC mikroaerobos apstākļos. Tomēr citohroma c peroksidāzes un NADH dehidrogenāzes negatīvajam dubultmutantam augšana 42oC aerobos apstākļos ir samazināta, salīdzinot ar NADH dehidrogenāzes negatīvo mutantu. Citohroma c peroksidāzes pārekspresija izraisīja ievērojamu skābekļa patēriņa samazināšanos, kas norāda uz jaunu, vēl nezināmu regulatoro efektu. NADH dehidrogenāzes negatīvajām mutantam bija paaugstināta termotolerance, bet laktātdehidrogenāzes un NADH dehidrogenāzes negatīvajā dubultmutantā šis efekts neizpaudās. Bakalaura darbs izstrādāts Latvijas Universitātes Mikrobioloģijas un Biotehnoloģijas institūtā. Zymomonas mobilis strains Zm6 and eight of its respiratory mutant derivates were grown in four different conditions: 30oC aerobic (in shaken flasks), 30oC microaerobic (without shaking), 42oC aerobic and 42oC microaerobic, to compare the oxygen and thermal stress impact on each of these strains. Strain growth, ethanol production and oxygen consumption were monitored. Overexpression of cytochrome c peroxidase improved growth at 30oC under microaerobic conditions. However, cytochrome c peroxidase and NADH dehydrogenase double knock-out strain showed impaired growth under 42oC aerobic conditions, when compared to the NADH dehydrogenase single knock-out. Unexpectedly, overexpression of cytochrome c peroxidase highly reduced oxygen consumption rate, pointing to yet unknown regulatory effects. NADH dehydrogenase knock-out mutation elevated thermotolerance, but knocking out the respiratory lactate dehydrogenase against the NADH dehydrogenase-deficient background reduced this effect.
