Peptīdu lunasīna un RGD regulējošā darbība peļu šizofrēnijas modeļos

Lunasīns ir bioloģiski aktīvs peptīds, kam piemīt antioksidatīvas, pretiekaisuma, pretvēža un holesterīna biosintēzi regulējošas īpašības. Nesen atklāts, ka tas spēj ietekmēt arī centrālo nervu sistēmu, izmainot peļu uzvedību, tādējādi radās interese pētīt lunasīna un tā sastāvā esošā tripeptīda RGD centrālos efektus. Maģistra darba mērķis bija pētīt intranazāli ievadītu peptīdu lunasīna un RGD darbību peļu šizofrēnijas modeļos, izmantojot atvērtā lauka testu un nosakot VMAT2 ekspresiju ar Western blot metodi. Iegūtie rezultāti rāda, ka gan lunasīns, gan RGD samazina amfetamīna un DOI izraisīto peļu horizontālo lokomotoro hiperaktivitāti, bet neietekmē fenciklidīna izraisīto vispārējo horizontālo lokomotoro hiperaktivitāti atvērtā lauka testā. Peļu smadzeņu corpus striatum netika konstatēta VMAT2 proteīna ekspresija. Lunasin is a bioactive peptide that has antioxidative, anti-inflammatory and others properties. It was recently discovered that lunasin can also affect the central nervous system, thus causing interest in studying the central effects of lunasin and tripeptide RGD. The aim of the master`s thesis was to study the activity of lunasin and RGD after intranasal administration in mice models of schizophrenia using the open field test and determining the expression of VMAT2 using the Western blot method. The results show that lunasin and RGD reduce the horizontal locomotor hyperactivity of mice induced by amphetamine and DOI, but does not affect the overall horizontal locomotor hiperactivity induced by phencyclidine in the open field test. In the corpus striatum of mice brain was not detected the expression of VMAT2 protein.
