Darba mērķis ir identificēt un raksturot gēnus, kas iesaistīti hipersensitīvās atbildes regulācijā miežos, kā modeļsistēmu izmantojot miežu nekrotiskos mutantus. Pētījums ietver divu iepriekš zināmu miežu nekrotisko mutantu – nec1 un nec3 – raksturošanu, kā arī iepriekš nezināmu anti-apoptotiskā slimību-izturības regulatora AtLSD1 miežu homologu identificēšanu un raksturošanu.
Pētījums balstīts uz kompleksu metodisko pieeja, kas apvieno gan tiešās ģenētikas (pozicionālo klonēšanu, uz transkriptomu balstīto klonēšanu), gan reversās ģenētikas metožu (TILLING populāciju pārbaudi, A. thaliana komplementāciju ar miežu gēniem), kā arī augu fizioloģijas un augu patoloģijas metožu izmantošanu. Pētījuma rezultāti atklāj iepriekš nezināmus miežu slimību-izturības aspektus.

Two known lesion mimic mutants – nec1 and nec3 – are chosen for the study. In addition, identification and characterization of previously unknown barley homologues of anti-apoptotic disease resistance regulator AtLSD1 is included in the study.
Study is based on integrated methodological approach, involving application of forward (positional cloning, transcript-based cloning) and reverse (TILLING population screening, complementation of model-organism A. thaliana with barley genes) genetics, as well as physiology and plant pathology – based experimental methods. Results reveal previously unknown aspects of barley disease resistance.
