Cilvēka 14.hromosomas proteasomu gēnu polimorfismu saistība ar metaboliskām un autoimūnām slimībām

Postgenoma ēras iestāšanās 21.gadsimtā izraisīja interesi par multifaktoriālo slimību ģenētiskajiem aspektiem, iespējamām saistībām starp kandidātgēnu alēļu variācijām un hroniskajām slimībām. Mēs savos pētījumos esam pievērsuši uzmanību 2.tipa cukura diabētam (T2DM) un juvenīlajam idiopātiskais artrītam (JIA). Asociācijas pētījumiem ar abām slimībām tika izvēlēti vairāki 14q hromosomu lokusi.
Cilvēka 14. hromosoma ir viena no piecām akrocentriskajām hromosomām, kurā ir lokalizēti astoņi proteasomu proteīnu gēni, tai skaitā, PSMB5, PSMB11, PSME1 un PSME2 lokusā 14q11.2, PSMA6 lokusā 14q13.1, PSMC6 lokusā 14q21.1, PSMA3 lokusā 14q23 un PSMC1 lokusā 14q32.11.
Darba mērķis bija raksturot saistību starp 14q polimorfismiem un JIA un T2DM Latvijas populācijā.
Piecus HSMS mikrosatelītu alēles un septiņus SNP (PSMB5 gēna SNP c.70C>T, PSMA6 gēna SNP c.-4543_-4544insC, c.-110C>A un c.-8C>G, PSMC6 gēna polimorfismu c.86-104A>G un c.86-46C>T un c.543+138G>A lokalizētu PSMA3 gēnā) genotipējām divos gadījuma/kontroles pētījumos, attiecīgi ar T2DM un JIA.
Sekvencējot noteicām, ka Latvijas populācijā HSMS602 marķiera motīvā ir divas mainīgas daļas jeb (CAA)n(A)m, HSMS701 ir ar vienkāršu atkārtojumu (AC)n.
Konstatējām, ka Latvijas populācijā T2DM ir saistīts ar HSMS006 alēli (TG)22, HSMS801 alēli (AC)24 un HSMS602 alēli (CAA)8, kā arī ar HSMS602 homozigoto genotipu (CAA)8/8 un HSMS801 heterozigoto genotipu (CA)20/22. Analizējot proteasomu proteīnu gēnu SNP, pierādījām, ka PSMA6 gēna c.-8C>G genotipu sadale ir saistīta ar T2DM Latvijas populācijā.
Ar JIA kopumā Latvijas populācijā ir saistītas HSMS006 alēles (TG)19 un (TG)23, HSMS701 alēle (AC)20 un HSMS801 alēle (AC)17, bet HSMS006 alēle (TG)20, HSMS702 alēles (TG)10 un HSMS602 alēles ar 178 bp ir saistīta ar JIoA un HSMS702 marķiera alēle (TG)13 - ar JIpA. Turklāt vairāki iepriekš minēto mikrosatelītu genotipi ir saistīti ar JIA Latvijas populācijā. SNP analīze uzrādīja, ka PSMA6 gēna c.-4543_-4544insC un c.-110C>A un PSMC6 gēna c.86-104A>G alēļu sadale ir saistīta ar JIA un tā vieglāko formu - oligoartītu. Turklāt c.86-104A>G retā alēle ir riska alēle oligoartrīta attīstībā (OR = 2,43). SNP c.-110C>A un c.86-104A>G genotipu sadale ir saistīta ar JIA un JIoA, bet PSMA3 gēna SNP c.543-138G>A genotipu sadale - ar JIA un JIpA. Advent of the post-genome era in the 21st century switched interest of many researchers to study of genetic component of multifactorial diseases, possible association of allelic variants of candidate genes with different chronic diseases are intensively sought. We have focused our attention on type 2 diabetes mellitus (T2DM) and juvenile idiopathic arthritis (JIA). The single 14q chromosome locus was chosen for association studies with both diseases.
Human chromosome 14 is one of five acrocentric chromosomes, harbouring eight proteasomal genes, including, PSMB5, PSMB11, PSME1 and PSME2 in locus 14q11.2, PSMA6 in locus 14q13.1, PSMC6 in locus 14q21.1, PSMA3 in locus 14q23 and PSMC1 in locus 14q32.11.
Goal of the study was to characterize association of 14q polymorphisms with T2DM and JIA in the Latvian population.
Five HSMS markers and seven SNPs (c.70C>T of PSMB5, c.-4543_-4544insC, c.110C>A and c.-8C>G of PSMA6, c.86-104A>G and c.86-46C>T of PSMC6 and PSMA3 SNP c.543+138G>A) were genotyped in two case/control studies with T2DM and JIA.
Direct sequencing revealed two variable components of the (CAA)n(A)m motif in HSMS602 marker and simple repeat motive (AC)n of the HSMS701 in Latvian population.
It was revealed that alleles (TG)22 of HSMS006, (AC)24 of HSMS801 and (CAA)8 of HSMS602 and also homozygote genotype (CAA)8/8 of HSMS602 and heterozygote genotype (CA)20/22 are associated with T2DM in Latvian population. By analyzing SNP of proteasomal genes, we have proved that genotypes distribution of c.-8C>G of PSMA6 gene is associated with T2DM in Latvian population.
Alleles (TG)19 and (TG)23 of HSMS006, (AC)20 of HSMS701 and (AC)17 of HSMS801 are associated with JIA in Latvian population, but alleles (TG)20 of HSMS006, (TG)10 of HSMS702 and 178bp of HSMS602 – with JIoA, and (TG)13 of HSMS702 – with JIpA. Also several genotypes of all microsatellites are associated with JIA in Latvian population. In study of SNP were develop that distribution of alleles of c.-4543_-4544insC and c.-110C>A of PSMA6 and PSMC6 SNP c.86-104A>G are relevant to JIA and JIoA, besides rear allele of c.86-104A>G is risk factor of JIoA (OR = 2.43). Genotype distribution of SNPs c.-110C>A and c.86-104A>G are associated with JIA and JIoA, but c.543-138G>A of PSMA3 – with JIA and JIpA.
