Stilbēna un čalkona sintāžu gēnu identifikācija krūmmellenes (Vaccinium corymbosum) genomā un sgRNS izveide inaktivācijai in vitro ar CRISPR/Cas9 metodi

Krūmmellenes (Vaccinium corymbosum L.) ir sastopamas pa visu Mērenā klimata joslu - gan Eiropas ziemeļos, gan Ziemeļamerikā. Pēdējos 20 gados vairāku iemeslu dēļ tās ar vien vairāk tiek kultivētas arī citos kontinentos. Tās ir viegli kultivēt, pateicoties tās labajai adaptācijas spējai pie temperatūras izmaiņām, tām ir izcila ogu kvalitāte, apjomīga raža un ievērojami liels sintezēto sekundāro metabolītu daudzums ogās. Tāpat kā vīnogās arī mellenēs tiek sintezēti ļoti daudz sekundāro metabolītu, pazīstamākā grupa ir antocianīni, kuri uzlabo cilvēku imūnsistēmas darbību. Lai gan mellenes aizvien vairāk tiek pētītas, jo ir ļoti nozīmīgs kultūraugs, šāds pētījums ir līdz šim veikts tikai ar vīnogām, kas pēc galvenā pētījuma objekta – antocianīnu sintāzēm - ir ļoti līdzīgas krūmmellenēm. Pētījuma galvenais mērķis bija izveidot sgRNS, kas varētu šķelt čalkona vai stilbēna sintāzes sekvenci krūmmellenes genomā un kuru varētu izmantot CRISPR/Cas9 sistēmā, lai inhibētu konkrēto sintāzi. Tādejādi resursi kļūs vairāk pieejami otrai sintāzei. Krūmmelleņu šķirņu ‘Duke’ un ‘Bluecrop’ genomos tika identificētas homologas čalkona un stilbēna sekvences, kas saturēja raksturīgās aminoskābju sekvences. Balstoties uz šīm homologajām sekvencēm, tika izveidotas sgRNS. Divi no četriem izveidotajiem sgRNS un Cas9 nukleāzes kompleksiem bija spējīgi daļēji sašķelt amplificēto DNS. Viens komplekss sašķēla šķirnes ‘Duke’ un otrs - ‘Bluecrop’ DNS. Atslēgas vārdi: krūmmellene, tetraploīds organisms, sekundārie metabolīti, antocianīni, bioķīmiskie signālceļi. Highbush blueberries (Vaccinium corymbosum L.) can be found all throughout the temperate zone, both in northern Europe and North America. Over the last 20 years they have been increasingly cultivated on other continents for a variety of reasons. They are easy to cultivate due to their ability to adapt to changes in temperature, they have excellent berry quality, a large yield and a considerably large amount of synthesized secondary metabolites, from which the best-known group is anthocyanins, which improve the functioning of the human immune system. Although blueberries are being studied more and more, because they are a very important crop, such a study has been carried out only on grapes, which are very similar to highbush blueberries according to the main object of the research - anthocyanin syntheses. The main objective of the study was to generate sgRNA that could chalcone or stilbene synthase sequence in the highbush blueberry genome, which could be used in the CRISPR / Cas9 system to inhibit the synthase. This will make resources more available for the second synthase. Homologous chalcone and stilbene sequences containing characteristic amino acid sequences were identified in highbush blueberries. Based on these homologous sequences, sgRNAs were generated. Two of the four created sgRNA and Cas9 nuclease complexes were able to partially split the amplified DNA. One complex split the DNA of ‘Duke’ and the other of ‘Bluecrop’ varieties.
