"Zymomonas mobilis elpošanas ķēdes loma šūnu dzīvotspējas saglabāšanā substrāta limita apstākļos"

Baktēriju elpošanas ķēdes (ETĶ) modifikācijas izmanto metaboliskajā inženierijā, tādēļ nepieciešams izprast ETĶ fizioloģiskās funkcijas. Šajā darbā tika salīdzināta Zymomonas mobilis ETĶ mutantu celmu un izejas celma Zm6 šūnu dzīvotspēja dažādos badināšanas režīmos – bez substrāta piedevām un ar glikozes vai etanola piedevām, kā arī iedarbojoties ar oksidatīvās fosforilēšanas (OF) inhibitoriem. Aerobi vislabāk auga NADH dehidrogenāzes (ndh) negatīvais celms Zm6-ndh. Badinot sešas stundas ar regulārām glikozes piedevām celmu dzīvotspēja bija augstāka, nekā badinot bez piedevām. Badinot ar etanola piedevām, augstāka dzīvotspēja bija tikai Zm6 un ar ndh komplementētajam Zm6-ndh_pNdh. OF inhibitori tiem samazināja dzīvotspēju. Tika secināts, ka etanola pievienošanas pozitīvais efekts ir saistīts ar ATF ģenerēšanu oksidatīvajā fosforilēšanā ETĶ. The name of this study is “The role of the respiratory chain of Zymomonas mobilis in the maintenance of cell viability under conditions of substrate limitation”. Modifications of the bacterial respiratory chain (ETC) are used in metabolic engineering, so it is necessary to understand the physiological functions of the ETC. In this work, the cell viability of Zymomonas mobilis ETC mutant strains and the original strain Zm6 was compared in different starvation regimes – without substrate additives, with glucose or ethanol additives, as well as exposure to oxidative phosphorylation (OF) inhibitors. The NADH dehydrogenase (ndh)-negative strain Zm6-ndh had the best aerobic growth. After starving for six hours with regular glucose supplementation, the viability of the strains was higher than when fasting without supplementation. When starved with ethanol additives, only Zm6 and ndh-complemented Zm6-ndh_pNdh had higher viability. OF inhibitors reduced their viability. It was concluded that the positive effect of adding ethanol is due to the generation of ATP by oxidative phosphorylation of ETC.
