MXene biomateriāls kā jauns pretvēža līdzeklis: šūnu mijiedarbības raksturojums

MXene ir plaši pētītu nanomateriālu grupa, kas tiek pētīta visdažādākajās jomās, tai skaita medicīnā, liekot uzsvaru uz pretvēža un antibakteriālo terapiju. Maģistra darbs tika fokusēts uz Ti3C2Tx MXene materiāla in vitro citotoksicitātes pētījumiem, uz kuriem balstās tālākie in vivo un in vitro pētījumi pretvēža fototermālajā terapijā. Tāpat tika analizēta MXene ietekme uz šūnu dzīvotspēju proteīnu klātbūtnē. Darba mērķis bija noteikt inovatīva MXene biomateriāla kā melanomas fototermālās terapijas kandidāta biosaderību ar epidermas un melanomas šūnu kultūrām in vitro. Šūnu dzīvotspēja tika analizēta izmantojot kolorimetrisko resazurīna metodi, bet MXene stabilitāte šķīdumā tika raksturota ar zeta potenciālu. Rezultāti uzrādīja, ka kerationcītu šūnu līnija ir daudz jutīgāka pret MXene iedarbību nekā melanomas šūnas, turklāt toksiskais efekts pret melanomas šūnām izpaužas tikai gadījumā, ja MXene iepriekš nav bijuši saskarē ar proteīniem. MXenes are a group of widely studied nanomaterials that are being studied in many fields, including medicine, with an emphasis on anticancer and antibacterial therapy. The Master's thesis was focused on the in vitro cytotoxicity studies of the Ti3C2Tx MXene, on which further in vivo and in vitro studies in anticancer photothermal therapy are based. The effect of MXenes on cell viability was also analyzed in the presence of protein coronas. The aim of the thesis was to determine the biocompatibility of the innovative MXene biomaterial as a candidate for melanoma photothermal therapy with epidermal and melanoma cell cultures in vitro. Cell viability was analyzed using the colorimetric resazurin assay, while the stability of MXene in solution was characterized by zeta potential. The results showed that the keratinocyte cell line is much more sensitive to the effects of MXenes than the melanoma cells, and the toxic effect against the melanoma cells is only present if MXenes have not previously been in contact with the proteins.
