Maģistra darbā ir pētīta mikrodaļiņu un nanodaļiņu (Ag, SiO2, Al2O3 un sukcinīta) dažādās ietekmes uz augu gametiskajām šūnām. Darba mērķis bija izpētīt šo daļiņu īstermiņa ietekmi uz augu gametiskajām šūnām. Kā modeļorganismi tika izmantoti miežu un alpu vijolīšu nenobriedušas mikrosporas. Izmantotās metodes bija plūsmas citometrija relatīvās fluorescences izmaiņu noteikšanai un DNS izdalīšana no miežu putekšņu šūnām. Retrotranspozonu izmaiņu noteikšanai tika izmantota iPBS metode. Kopumā Al2O3 un SiO2 mikrodaļiņām ir negatīva ietekme uz mikrosporu pašfluorescenci, bet sukcinīta (dzintara) daļiņas to paaugstina, bet Ag daļiņām šajā pētījumā nav novērota ietekme uz mikrosporām. iPBS metode uzrādīja izmaiņas miežu mikrosporu DNS SiO2, Al2O3 un sukcinītā inkubētajos paraugos. The master's thesis focus was studying the different effects of microparticles and nanoparticles (Ag, SiO2, Al2O3 and succinite) on plant gametic cells. The aim of the work was to investigate the short-term effects of these particles on plant gametic cells. Immature microspores of barley and alpine violets were used as model organisms. The methods used were flow cytometry for relative fluorescence changes and DNA extraction from barley pollen cells. The iPBS method was used to detect retrotransposon changes. In general, Al2O3 and SiO2 microparticles have a negative effect on the autofluorescence of microspores, while succinite (amber) particles increase it, and Ag particles have no effect on microspores in this study. The iPBS method showed changes in barley microspore DNA in SiO2, Al2O3 and succinite incubated samples.
