Zymomonas mobilis elpošanas ķēdes regulācijas izpēte stresa apstākļos izmantojot zaļā fluorescentā proteīna reportiersistēmu

Zymomonas mobilis ir etanologēna baktērija ar potenciālu pielietojumu biodegvielas ražošanā. Lai izpētītu elpošanas ķēdes lomu pie etanola un sāls stresa apstākļiem tika izveidota reportiergēnu sistēma ar elpošanas ķēdes komponentu promoteriem un EGFP. Iegūtie dati liecina ka visspēcīgākais promoters ir II tipa NADH dehidrogenāzei (Ndh). Pārējo apskatīto komponentu citohroma bd, citohroma bc1 un citohroma c peroksidāzes promoteru aktivitāte bija vāja. Pie stresa visu promoteru aktivitāte samazinājās, ar izteiktāku efektu pie sāls stresa, kas varētu būt saistīts ar oksidatīvā pēcstresa samazināšanu. Ndh enzīma aktivitāte pie sāls stresa pieauga. Tas varētu būt saistīts ar post-translācijas regulāciju. Zm6- ndh pie sāls stresa aug labāk par Zm6, kas liecina ka Ndh aktivitāte neuzlabo sāls stresa toleranci. Zymomonas mobilis is an ethanologenic bacterium with potential applications in biofuel production. To investigate the role of the respiratory chain under ethanol and salt stress conditions, a reporter gene system with respiratory chain protein promoters and EGFP was created. Results show that the strongest promoter is type II NADH dehydrogenase (Ndh). Activity of cytochrome bd, cytochrome bc1 and cytochrome c peroxidase promoters was weak. Under stress, the activity of all promoters decreased, with a more pronounced effect under salt stress, which could be related to the reduction of oxidative post-stress. Ndh enzyme activity increased under salt stress. This could be due to post-transcription regulation. Zm6-ndh strain grows better than Zm6 under salt stress, which indicates that Ndh activity does not improve salt stress tolerance.
