Daudzgadīgās airenes Lolium perenne abiotiskā stresa tolerances kandidātgēna CBP60G rediģēšanai paredzētu plazmīdu konstrukciju sagatavošana un funkcionalitātes pārbaude

Darba mērķis bija sagatavot un pārbaudīt plazmīdu konstrukcijas L.perenne CBP60G kandidātgēnu rediģēšanai ar CRISPR/Cas9 metodi. In silico tika identificēti AtCBP60g gēna L. perenne homologi un veikta šo gēnu sekvencēšana no L.perenne selekcijas līnijām un cv ‘Veja’. Balstoties uz iegūtām sekvencēm tika izvēlētas mērķa sekvences (protospacer) mutāciju ieviešanai ar CRISPR/Cas9 metodi, kā arī izveidoti attiecīgās gRNA saturoši konstrukti protoplastu transformācijai. Konstruktu genoma rediģēšanas efektivitāte protoplastos tika pārbaudīta ar 2 metodēm: ar Sangera sekvencēšanu un heterodupleksu analīzi. Rezultātā tika konstatēti astoņi AtCBP60G homologi ar zemu sekvences līdzību A.thaliana proteīnam, no kuriem trim tika sagatavoti gRNA saturoši konstrukti mutāciju ieviešanai. No trim rediģētiem kandidātgēniem sekmīga rediģēšana tika panākta divos. Kandidātgēnu rediģēšanas sekmes ar izveidotajiem gRNA konstruktiem bija salīdzinoši zemas, tomēr rezultāti apstiprina izvēlētās metodiskās pieejas pielietojamību L.perenne genoma rediģēšanai ar CRISPR/Cas9 metodi. The aim of this work was to prepare and test plasmid constructs for editing L. perenne CBP60G candidate genes by CRISPR/Cas9. Based on sequence homology, eight candidate loci were identified in L. perenne. These loci were sequenced from L.perenne breeding lines and cv ‘Veja’. The obtained sequences were used for selection of protospacers for gene-specific mutagenesis using CRISPR/Cas9 approach. gRNA constructs for gene editing were prepared for three of homologous loci. Gene editing efficiency of the developed gRNA constructs was tested using two approaches – Sanger sequencing and heteroduplex analysis. As a result, eight homologous candidate genes from L.perenne were identified. They shared low sequence homology with the AtCBP60g. Out of three gRNA constructs tested for gene editing, two were successful in the editing of target genes. Although overall editing efficiency was low, results confirm the applicability of the applied approach for CRISPR/Cas9-based editing of L.perenne genome.
