Biomaterialu testēšana uz citotoksitati un funkcionalitāti

Biomateriālu izmantošana pēdējos gados ir kļuvusi par neatņemamu reģeneratīvās medicīnas sastāvdaļu, kur tie tiek izmantoti, lai aizstātu bojātos audus un veicinātu audu vai orgānu atjaunošanu. Kaulaudu atjaunošanā biomateriālu loma ir nodrošināt struktūru, kura veicina priekštečšūnu migrāciju, dalīšanos un diferencēšanos. Jaunu biomateriālu izstrādē būtiski ir novērtēt to drošību un spēju veicināt kaulaudu reģenerāciju. Kā viens no pirmajiem šādas izstrādes posmiem ir laboratoriska, in vitro, citotoksicitātes un biosaderības novērtēšana. Bakalaura darba ietvaros veikta divu jaunizstrādātu hidrogelu tipa biomateriālu in vitro novērtēšana. Izmantojot dažādas metodes, tika analizēta biomateriālu ietekme uz šūnu dzīvotspēju un proliferāciju MG-63 un MC3T3-E1 šūnu līnijās. Papildus tika analizētas osteinduktīvās īpašības testējot biomateriālu ietekmi uz pre-kolagēna I un matriksa metalloproteināzes 1 (MMP1) sekrēciju. Rezultāti ietver biomateriālu biosaderības raksturojumu. Tie parāda atšķirīga sastāva biomateriālu atšķrīgo ietekemi uz šūnu dzīvotspēju, dalīšanās aktivitāti, prekolagēna I un MMP1 sekrēciju. In recent years, the use of biomaterials has become an integral part of regenerative medicine, where they are used to improve tissue and promote tissue or organ regeneration. In the regeneration of bone tissue, the role of biomaterials is to provide a structure that promotes the migration, division and differentiation of progenitor cells. In the development of new biomaterials, safety and the ability to promote bone tissue regeneration have been evaluated. One of the first steps in such development is laboratory, in vitro, cytotoxicity and biocompatibility evaluation. In vitro evaluation of two newly developed hydrogel-type biomaterials was performed as part of the bachelor thesis. The effects of biomaterials on cell viability and proliferation in MG-63 and MC3T3-E1 cell lines have only been analyzed using different methods. In addition, osteoinductive properties were analyzed by testing the effect of biomaterials on pre-collagen I and matrix metalloproteinase 1 (MMP1) secretion. The results include biocompatibility characterization of biomaterials. They show the different effects of biomaterials of different composition on cell viability, division activity, precollagen I and MMP1 secretion.
