Savienojuma X un R-fenotropila antidepresanta aktivitāte žurkām

Abstract

Psihiskas slimības, tai skaitā depresija, skar trešo daļu Eiropas iedzīvotāju un Latvijā - ap 120 000 cilvēku. Jau 2020.gadā varētu izveidoties situācija, ka depresija ir viens no galvenajiem darba nespējas cēloņiem pasaulē. Pirmo reizi pēc hroniskas vielu ievadīšanas tika pārbaudīta jauna savienojuma X antidepresanta aktivitāte žurkām piespiedu peldēšanas testā divās sērijās. Pirmajā sērijā vielas ievadītas intraperitoneāli intaktām žurkām devās 1mg/kg, 10mg/kg un 25mg/kg, kā references vielu izmantojot R-fenotropilu 10mg/kg. Otrajā sērijā vielas ievadītas intraperitoneāli žurkām, kas iepriekš veikušas atmiņas testu, devās 1mg/kg, 5mg/kg un 10mg/kg, kā references vielu izmantojot R-fenotropilu devās 1mg/kg un 10mg/kg. Pētījuma rezultāti parāda, ka savienojums X neuzrāda antidepresanta aktivitāti. Netika novērota statistiski būtiska atšķirība ne ar kontroles vielu, ne ar references vielu R-fenotropilu nevienā no eksperimenta sērijām.

Mental disorders, including depression affect about one third of European population. In Latvia, 120000 patients are suffering from depression. It is suggested that already in 2020, the main cause of work disability all over the world will be depressive diseases. The present work studied the novel compound X to assess its antidepressant activity in rats. R-phenotropil was selected as a reference drug, saline for control. The forced swimming test was used. The experiments were carried out by use of two schedules. In one, the compound X and R-phenotropil were injected intraperitoneally to intact rats at doses 1, 10 and 25mg/kg for 10 days. In the other experimental series, rats prior to swimming test had undergone a memory test; the compound X was administered at doses 1, 5 and 10mg/kg, and R-phenotropil at 1 and 10 mg/kg , both substances were injected for 10 days. The obtained data demonstrated that neither compound X nor R-phenotropil possessed antidepressant activity.
