ds RNS kvantitatīvās noteikšanas metodes izstrāde rektālai zāļu formai "Larifāns"

Abstract

Maģistra darba mērķis bija izstrādāt jaunu metodi, lai noteiktu dubultspirāliskās ribonukleīnskābes (ds RNS) kvantitatīvo sastāvu rektālajā zāļu formā „Larifan Supo” ar devām 0,5 mg, 1,0 mg un 1,5 mg. Dubultspirāliskās RNS noteikšana tika veikta ar jonu apmaiņas apgrieztas fāzes augstefektīvo šķidruma hromatogrāfiju (AEŠH). Rezultātā tika izstrādāta jauna metode ds RNS kvantitatīvai noteikšanai, kura validēta, saskaņā ar ICH Q2B vadlīnijām. Pēc veiktās analīzes, ir noskaidrots, ka metode ir lineāra, precīza un specifiska, un atbilstoša ds RNS kvantitatīvajai analīzei. Analizējamie paraugi istabas temperatūrā 24 stundu periodā ir stabili, un paraugi šajā periodā dod atkārtojamus un precīzus rezultātus.

The aim of the research was to develop a new method of quantitative determination of double-stranded ribonucleic acid (dsRNA) in rectal suppositories „Larifan Supo” with dosages 0,5 mg, 1,0 mg and 1,5 mg. Double-stranded RNA was determinated with ion-exchange reversed phase high performance liquid chromatography (HPLC). The new method of quantitative determination was developed and afterwards validated by ICH Q2B guidelines. After analysis, results verified that method is linear, precise, specific and appropriate for quantitative determination of dsRNA. Solution samples that were used for analasys, were stable at room temperature for 24 hours, and in this time period can give repeatable and highly precise results.
