Zema melanomas riska gēnu polimorfismi Latvijas populācijā

Melanoma ir ļaundabīgs ādas audzējs, kas attīstās no melanocītiem. Visvairāk pētītais zema melanomas riska gēns ir melanokortīna 1. receptora gēns (MC1R). Vairāki tā polimorfismi saistīti ar melanomas risku. No citiem melanomas riska polimorfismiem līdz šim visaugstāko saistību ir uzrādījuši polimorfismi rs1393350 (11q14-q21), rs258322, rs4785763 un rs8059973 (16q24). Maģistra darba mērķis bija izvērtēt zema melanomas riska gēnu polimorfismu ietekmi uz melanomas attīstību Latvijā. MC1R gēna polimorfismu analīze veikta izmantojot PCR un sekvenēšanu. Četri atlasītie polimorfismi genotipēti izmantojot Q-PCR. MC1R gēna polimorfismi izanalizēti 200 melanomas pacientiem un 200 kontroles personām. Polimorfismi konstatēti 79,5% melanomas pacientu un 62,5% kontroles personu. Kopumā tika atrasti 26 dažādi polimorfismi. Individuāli Latvijas populācijā ar melanomas risku saistīti polimorfismi Val60Leu, Val92Met, Arg151Cys un Arg160Trp, kuru gadījumā risks paaugstinās 2 - 4 reizes. Pēc meta analīzes risku saglabā Val60Leu, Arg151Cys un Arg160Trp. Latvijas populācijā melanomas pacientiem raksturīgs melanomas riska fenotips - gaiša āda un mati. Individuāli asociāciju ar augsta riska fenotipu uzrāda polimorfismi Val92Met, Arg151Cys un Arg160Trp. Stratificējot personas pēc pigmentācijas fenotipa, polimorfismi paaugstina melanomas risku personām ar zema melanomas riska fenotipu - tumšu ādu un matiem. Tas liecina, ka Latvijas populācijā MC1R gēna polimorfismi ietekmē melanomas risku neatkarīgi no pigmentācijas fenotipa. Četru riska polimorfismu analīze parādīja, ka ar melanomu saistīti 16. hromosomas polimorfismi rs258322 un rs4785763, kas atrodas vienā haplotipā ar MC1R gēna polimorfismiem Val92Met, Arg151Cys un Arg160Trp. Maģistra darbs izstrādāts Latvijas Biomedicīnas pētījumu un studiju centrā laika posmā no 2009. gada septembra līdz 2011. gada maijam. Atslēgas vārdi: melanoma, melanokortīna 1. receptora gēns, MC1R polimorfismi. Cutaneous melanoma is malignant skin cancer, that develops from melanocytes. The most studied low melanoma risk susceptibility gene is melanocortin 1 receptor gene (MC1R). Some of these polymorphisms are associated with melanoma. In terms of other melanoma risk polymorphisms, rs1393350 (11q14-q21), rs258322, rs4785763, and rs8059973 (16q24) have presented the highest association so far. The aim of the master’s thesis was to evaluate the effect of low melanoma risk polymorphisms on melanoma development in Latvia. The analysis of MC1R gene polymorphisms was done by PCR and sequencing. Four selected melanoma risk polymorphisms were genotyped by Q-PCR. MC1R gene polymorphisms were studied in 200 melanoma patients and 200 control persons. Polymorphisms were found in 79.5% melanoma patients and 62.5% control persons. Altogether 26 polymorphisms were identified. Individually the risk is associated with polymorphisms Val60Leu, Val92Met, Arg151Cys and Arg160Trp, that increases risk 2 - 4 times. After meta-analysis Val60Leu, Arg151Cys and Arg160Trp preserves the risk. Melanoma patients in Latvian population carry high melanoma risk phenotype - light skin and hair. Individually an association with the high risk phenotype present Val92Met, Arg151Cys and Arg160Trp. After stratification by pigmentation phenotype, polymorphisms increase melanoma risk in persons with low risk phenotype - dark skin and hair. It indicates, that in Latvian population MC1R gene polymorphisms influence melanoma risk independently of pigmentation phenotype. The analysis of four melanoma risk polymorphisms showed, that polymorphisms rs258322 and rs4785763 of chromosome 16 are associated with melanoma and they are in one haplotype with MC1R gene polymorphisms Val92Met, Arg151Cys and Arg160Trp. The research for the master thesis was carried out at the Latvian Biomedical Research and Study Centre from September 2009 till May 2011.
