dc.description.abstract | IS10 ir viens no aktīvākajiem transpozoniem baktērijās. Elementam ir liela nozīme Escherichia coli genomā pārkārtošanās, kas notiek šūnu kultūrā iestājoties stacionārai augšanas fāzei. Ģenētisko izmaiņu rezultātā veidojas adaptīvās mutācijas, kas pārsvarā ir neekspresējamo gēnu aktivācijas rezultāts. Darba mērķis bija noskaidrot IS10 izraisīto nefunkcionējoša hloramfenikola acetiltransferāzes gēna aktivācijas iespējamos mehānismus. Izpētīts, ka plazmīda pAdCat ir efektīvs vektors IS10 izoformas – IS10H secības izolēšanai. Modelēts, ka bezpromotera cat varētu būt aktivizēts ar iekšējām IS10H sekvencēm, kas veido baktēriju -10 un -35 promoteriem attāli homologas secības. Noteikts, kā IS10H integrācijas sekvences atšķiras no aprakstītajām IS10R elementam. Ar DNS blotu noteiktas būtiskās atšķirības IS10 izplatībā genomā diviem tuvu radniecīgiem XL1-blue celma atvasinājumiem. Noteikta pozitīvā korelācija starp starpreplikonu transpozīcijas biežumu un elementu skaitu genomā. Intragenomiskā elementu migrēšana netika konstatēta, arī pēc ģenētiskā un metaboliskā stresa izraisīšanas šūnām. | en_US |
dc.description.abstract | IS10 is one of the most active transposons in bacteria. This transposable element plays an important role in Escherichia coli genome rearrangements that occur in cell culture entering stationary growth phase. Genetic changes may take place in the form of adaptive mutations, through activation of the non-expressed genes.
The goal of this work was to clarify the mechanisms of the IS10-dependent activation of the promoterless chloramphenicol acetyltransferase (cat) gene. A plasmid pAdCat is an effective entrapment vector for IS10 isoform – IS10H isolation. The cat gene transcription could be triggered by internal IS10H sequences that are remotely homologous to the bacterial -10 and -35 promoter boxes. IS10H integration sites in pAdCat differ from the sites described for IS10R element. DNA blot showed prominent differences in the distribution of the IS10 sequences within closely related XL1-blue strains. We have established a positive correlation between genome to plasmid transposition frequency and number of elements in the genome. Destabilization of IS10 distribution pattern within the bacterial genome, an intragenomic element migration, was not observed even after inducing genetic and metabolic stress of the cells at prolonged maintenance in the stationary cultivation phase, when genome to plasmid transposition of IS10H occurs. | en_US |
