Bakalaura darba mērķis ir izveidot konstrukcijas, ar kuru palīdzību, izmantojot baktēriju divhibrīdu sistēmu, varēs identificēt RGMoV kodēto proteīnu savstarpējās mijiedarbības.
Bakalaura darbs tika izstrādāts no 2010.10. - 2011.05. BMC Rekombinanto proteīnu laboratorijā Dr. biol. Andra Zeltiņa un Dr. biol. Inas Baļķes vadībā, par pētījumu objektu izmantojot daudzziedu airenes raibuma vīrusu.
Bakalaura darbā tika izveidotas baktēriju divhibrīdu sistēmas konstrukcijas, kas satur visus identificētos RGMoV kodētos pilna garuma proteīnu gēnus gan „ēsmas”, gan mērķa vektoros un pārbaudīta to savstarpējā mijiedarbība.
Mijiedarbību pētījumos tika konstatēts, ka mijiedarbojas („ēsma”-mērķis) CP-CP, CP-P16, CP-RdRp, CP-SerP, P1-CP, P1-P16, P1-RdRp, P1-SerP, P16-CP, P16-P16, P16-RdRp, P16-SerP, RdRp-CP, RdRp-P16, RdRp-RdRp, RdRp-SerP, SerP-P16, SerP-RdRp, VPg-CP, VPg-P16 un VPg-RdRp.
Atslēgas vārdi: bakteriālā divhibrīdu sistēma, proteīnu mijiedarbības, RGMoV. The aim of this work is to create RGMoV protein encoding bacterial two hybrid system vectors for protein-protein interaction studies using the bacterial two-hybrid system.
This work has been worked out from October 2010 till May 2011 in BMC, Recombinant protein laboratory under the supervision of Dr. biol. Andris Zeltiņš and Dr. biol. Ina Baļķe. Subject of the study is the Ryegrass mottle virus.
Several bacterial two hybrid system constructs containing all of the full length so far identified RGMoV protein encoding sequences were made, each in the “bait” vector and target vector.
From the interaction experiments several interactions were detected: CP-CP, CP-P16, CP-RdRp, CP-SerP, P1-CP, P1-P16, P1-RdRp, P1-SerP, P16-CP, P16-P16, P16-RdRp, P16-SerP, RdRp-CP, RdRp-P16, RdRp-RdRp, RdRp-SerP, SerP-P16, SerP-RdRp, VPg-CP, VPg-P16 un VPg-RdRp..
