Dažādu E. coli serotipu attīstība maltajā gaļā.

Escherichia coli ir Gramnegatīva nesporulējoša nūjiņa, kas ir samērā noturīga pret ārējās vides apstākļiem, un vidē var saglabāties pat vairākus mēnešus, tāpēc Escherichia coli izmanto kā fekālā piesārņojuma indikator-mikroorganismu. Tai ir sarežģīta antigēnu struktūra, no kurām diagnosticēšanā nozīmīgākais ir somatiskais (O) antigēns un viciņu (H) antigēns.
Eiropas Komisijas regula (EK) Nr. 2073/2005 (2005.gada 15.novembris) par pārtikas produktu mikrobioloģiskajiem kritērijiem nosaka, ka Escherichia coli izmanto kā indikator-mikroorganismu, lai novērtētu ražošanas procesa vai produkta higiēnu. Testēšanas metode (ISO 16649-2), kura tiek norādīta šajā regulā, balstās uz E.coli spēju producēt fermentu β-glikuronidāzi, taču šī fermenta aktivitāte novērojama apmēram 97% no E.coli.
Pētījumā tika izmantoti trīs E.coli serotipi (O26, O103, 145), kuri visbiežāk sastopami pārtikas produktos. Kā matrice tika izmantota trīs dažādas maltās gaļas (malta liellopu gaļa, malta cūkgaļa un jaukta maltā gaļa (liellopu gaļa:cūkgaļa=1:1)). Matricām izvirzītās prasības bija, lai tās nesaturētu konservantus un E.coli.
Katrs maltās gaļas veids tika kontaminēts ar katru no E.coli serotipiem, un pētīta to attīstība +20C un +40C (temperatūras robežas kādās ražotāji iesaka uzglabāt malto gaļu) un +80C (temperatūra, kāda, pēc veiktajiem pētījumiem, ir sastopamas mājsaimniecību ledusskapjos).
Escherichia coli serotipi O26, O103 un O145 2-4°C temperatūrā vairojas lēni un pat pēc maltās gaļas 96 stundu ilgas uzglabāšanas šādās temperatūrās nepārsniedz Eiropas Komisijas regulas Nr.2073/2005 par pārtikas produktu mikrobioloģiskajiem kritērijiem minēto maksimāli pieļaujamo vērtību (500 KVV/g), taču E.coli serotipu O26, O103 un O145 attīstība 8°C temperatūrā notiek straujāk, un jau pēc 48 stundām vidēji trīs reizes pārsniedz maksimāli pieļaujamo E.coli skaitu maltajā gaļā.

European Commision regulation (EC) No 2073/2005 of November 2005 on microbiological criteria for foodstuffs lays down to use Escherichia coli to indicate fecal contamination for evaluation of hygiene at production area and products.
Method ISO 16649-2 mentioned in this EC regulation is based on ability of E.coli produce β –glucuronidase, but this ferment has only 97% of all E.coli.
In this research were used three serotypes of E.coli (O26, O103, 145) that are more frequently found in food products. Grounded beef, grounded pork and grounded mixed meat (beef meat:pork meat=1:1) was used as matrix. Absence of E.coli and absence of preservatives were the requirements for matrixes.
All grounded meats were contaminated with each serotype of E.coli and examinated differences during storage at +20C and +40C temperature (storage temperature defined by producers) and at +80C (temperature that were found to be in refrigerators at households).
Multiplication of Escherichia coli serotype O26, O103 un O145 at 2-4°C temperature is slow and after 96 hours storing at these temperatures, meet requirements (500 cfu/g) mentioned in European Commision regulation (EC) No 2073/2005 of November 2005 on microbiological criteria for foodstuffs. But multiplication of E.coli serotype O26, O103 and O145 at 8°C temperature are significant and after 48 hours exceed more than three time maximum allowed E.coli count in grounded meat.
