TP53, MDM2 un MC1R gēna polimorfismu saistība ar melanomas risku Latvijas populācijā

Melanoma ir melanocītu ļaundabīgs ādas audzējs. Zināmi tikai divi augstas penetrances melanomas jutības gēni (CDKN2A, CDK4), tāpēc tiek pētīta vairāku zemas penetrances melanomas jutības gēnu saistība ar melanomas risku.
Bakalaura darba mērķis bija izanalizēt trīs P53 signālceļa gēnu (TP53, MDM2 un MC1R) polimorfismu ietekmi uz melanomas risku Latvijas populācijā.
TP53 polimorfisms Pro72Arg tika izanalizēts 245 melanomas pacientiem un 194 kontroles personām. Polimorfismu c.14+309T>G saturošais MDM2 1. introna fragments tika nosekvenēts 259 melanomas pacientiem un 191 kontroles personai.
Statistiski ticama saistība ar melanomu tika konstatēta MDM2 polimorfisma c.14+309T>G G alēlei personām, kas sasniegušas vismaz 50 gadu vecumu (OR=1,61), kā arī T/G genotipam kombinācijā ar jebkuru nesinonīmo MC1R polimorfismu (OR=2,61). Statistiski ticama saistība novērota arī TP53 Pro72Arg G/G genotipam kombinācijā ar RHC, kā arī jebkuru nesinonīmo MC1R polimorfismu (attiecīgi OR=3,46 un 3,15). Melanoma is a malignant skin tumor of melanocytes. There are known only two high penetrance melanoma susceptibility genes (CDKN2A, CDK4), therefore some low-penetrance susceptibility genes are studied.
The objective of the Bachelor's thesis was to analyze the impact of polymorphisms in three P53 signaling pathway genes on melanoma risk in Latvian population.
TP53 polymorphism Pro72Arg was analyzed in 245 melanoma patients and 194 control individuals. MDM2 polymorphism c.14+309T>G was analysed in 259 melanoma patients and 191 control persons.
Statistically significant association with melanoma was detected in persons with G allele of MDM2 polymorphism c.14+309T>G, who are at least 50 years old (OR=1.61), as well as for T/G genotype in combination with any of MC1R polymorphisms (OR=2.61). Statistically significant association with melanoma was also detected in carriers of TP53 Pro72Arg G/G genotype in combination with RHC or any MC1R polymorphisms (OR=3.46 and 3.15, respectively).
