dc.description.abstract | Bakalaura darba mērķis ir pārbaudīt RGMoV poliproteīnu domēnus VPg un P16 kā iespējamos partnerus sajūgto proteīnu ekspresijas sistēmā. Par mērķa proteīniem izmantojot RGMoV RdRp, SerP un P1 proteīnus. Izveidotās konstrukcijas tika ekspresētas E. coli BL21(DE3) vai WK6 ekspresijas šūnu celmos.
Bakalaura darbs izstrādāts no 2011. gada februāra līdz 2011. gada maijam BMC Rekombinanto proteīnu laboratorijā Dr. biol. Andra Zeltiņa un Dr. biol. Inas Baļķes vadībā. Pētījumu objekts ir daudzziedu airenes raibuma vīruss.
Bakalaura darbā izveidotas konstrukcijas: pET-Du-P16-RdRp, pCold-P16-RdRp, pET-P1-VPg, pET-SerP-VPg, kas satur P16, VPg, P1, SerP un RdRp.
Iegūtie sajūgto proteīnu ekspresijas dati uzrādīja, ka VPg un P16 var kalpot kā potenciālie sajūgtie proteīni, jo SerP gadījumā tika iegūta salīdzinoši augsta ekspresija pET sistēmā, kas iepriekš netika panākta, kā arī RdRp neliela ekspresija pET-Duet sistēmā.

dc.description.abstract | The aim of this work is to test RGMoV poliprotein domain VPg and P16 as possible fusion partners in a fusion protein expression system. RGMoV RdRp, SerP and P1 proteins were used as fusion target proteins. Created constructs will be expressed in E.coli BL21(DE3) or WK6 lines.
Presented work was conducted from February 2011 till May 2011 in BMC, Recombinant protein laboratory under the supervision of Dr. biol. Andris Zeltiņš and Dr. biol. Ina Baļķe. Subject of the study is the ryegrass mottle virus.
During this work several expression plasmids were constructed: pET-Du-P16-RdRp, pCold-P16-RdRp, pET-P1-VPg, pET-SerP-VPg containing P16, VPg, P1, SerP and RdRp encoding sequences.
Protein expression experiments demonstrated that P16 and VPg are suitable as potential fusion partners. They had a positive effect on SerP expression in pET expression system, and also a smaller effect on RdRp expression in pET-Duet system.
