dc.description.abstractLentinula edodes ir plaši kultivējama sēne, kura sintezē enzīmus spējīgus degradēt koksnē esošo lignīnu. Šī pētījuma mērķis ir atlasīt Lentinula edodes celmus pēc to spējas sintezēt ligninolītiskos enzīmus un degradēt lignīnu. Vispirms tika veikts 15 L. edodes celmu skrīnings lignīnu degradējošo enzīmu notektēšanai, tad celmi tika audzēti barotnē ar un bez lignocelulozes (LC) piedevas un noteikta enzīma lakkāzes aktivitāte. Tika noteikts, ka visi L. edodes celmi sintezē divu veidu lignīnu degradējošos enzīmus – lakkāzi un mangāna peroksidāzi. Celmu augšanas intensitāte būtiski neatšķīrās, augot uz agarizētās barotnes ar un bez LC piedevas, bet celmu micēlija biomasa pieauga līdz pat 2 reizēm, augot šķidrajā barotnē ar LC piedevu. Enzīmus ar vilielāko aktivitāti sintezēja celmi 386, 395, 3565 un 4b2c.en_US
dc.description.abstractLentinula edodes is wide cultivated mushroom, which is able to synthesize enzymes to degrade lignin in the wood. Purpose of this study is to select L. edodes strains for their ability to synthesize ligninolytic enzymes and to degrade lignin. Firstly was made qualitative screening for ligninolytic enzymes activities of 15 L. edodes strains. Then strains were cultivated on solid media and in liquid media with lignicellulose (LC) additive and without it and was determined enzyme laccase activity with photocalorimetric methode. There was determined that all L. edodes strains synthesize two kinds of lignin degrading enzymes – laccase and manganese peroxidase. Micelial growth intensity of strains did not differ significantly, growing on solid media with LC adding and without it. But mycelial biomass of L. edodes strains increase more than two times, growing in liquid media with LC additive. Enzyme with the highest activity was synthesized by strains 386, 395, 3565 and 4b2c.en_US
