Darba mērķis bija panākt rekombinanta šķīstoša cilvēka CCR2 hemokīnu receptora analoga ekspresiju baktēriju šūnās.Rekombinantā šķīstošā CCR2 receptora (rsCCR2) kodējošais DNS fragments tika sintezēts mākslīgi. Konstrukcijā ietilpstošā aminoterminālā domēna, ekstracelulāro cilpu un tām blakusesošo transmembranālo domēnu fragmentu kodējošās sekvences tika savienotas, izmantojot mākslīgus DNS linkerus. rsCCR2 sintēzi baktēriju šūnās BL21 (DE3) izdevās panākt, izmantojot ekspresijas vektoru pCold1. Iegūtais nešķīstošais proteīns tika attīrīts ar metālafīno hromatogrāfiju un refoldēts ar iznākumu 1 mg/ml. Lai apstiprinātu iegūtā rekombinantā proteīna funkcionalitāti, tika izmantots imunoblots un ELISA. Imunoblota rezultāti liecina, ka komerciālas anti-CCR2 antivielas atpazīst rekombinanto proteīnu. Sekmīgai Sandwich ELISA sistēmas izveidei nepieciešama papildus optimizācija, lai panāktu specifisku rsCCR2 saistīšanos un kvantificēšanu. The aim of this investigation was to achieve the expression of recombinant soluble human CCR2 chemokine receptor in bacteria.The recombinant soluble CCR2 receptor (rsCCR2) coding DNA fragment was artificially synthesized, in which N-end domain was connected with two extracellular loops and adjacent fragments of transmembranal domains using artificial DNA linkers. It was possible to achieve rsCCR2 protein expression in bacteria cells BL21 (DE3) using expression vector pCold1. Obtained insoluble protein was purified using metal affinity chromatography and refolded with the yield ~1 mg/ml. To validate the functionality of the recombinant rsCCR2 Western Blot and ELISA were used. With Western Blot it was demonstrated that commercial anti-CCR2 antibodies are able to recognize N-terminus of rsCCR2. ELISA system needs to be optimized to obtain specific rsCCR2 binding and quantification.
