KOPSAVILKUMS
Maģistra darba mērķis bija salīdzināt metodes optimālas Phytophthora ramorum noteikšanas shēmas izstrādei.
Darbā pētītas septiņas kultivēšanas barotnes, četras DNS izdalīšanas metodes un divi PCR protokoli, kā arī divas rododendru lapu inficēšanas metodes.
Konstatēts, ka vispiemērotākā metode P.ramorum virulences saglabāšanai, ir auga materiāla inficēšana mitrajā kamerā. No darbā izmantotajām septiņām barotnēm V8-2 barotne ir visvairāk piemērota darbam ar P.ramorum, jo uz tās vislabāk attīstās sēnes sporangiji. Uz PARP/H, burkānu gabalu, V8-1, V8-3 un V8-4 barotnēm P.ramorum micēlija morfoloģija atbilst literatūrā aprakstītajai, bet uz V8-2 barotnes tas ir morfoloģiski atšķirīgs – ļoti plāns micēlijs un nav koraļļveidīgs. Identifikācijai būtiskākās struktūras – hlamidosporas uns sporangiji - morfoloģiski līdzīgi uz visām barotnēm.
Llop et al. (1999) metode ir vispiemērotākā P.ramorum DNS izdalīšanai no augu materiāla. DNS izdalīšanai no P.ramorum tīrkultūras piemērotākās ir Llop et al. (1999) un Qiagen DNeasy augu DNS izdalīšanas metodes, kas ļauj iegūt kvalitatīvāku DNS nekā pārējās darbā pētītās metodēs.
Ar Llop et al. (1999) un Qiagen DNeasy metodēm izdalītais DNS uzrāda līdzīgus rezultātus vienpakāpes un divpakāpju PCR. Comparison of different methods for optimization of Phytophthora ramorum detection scheme.
The goal of my research was to compare of different methods for optimization of P.ramorum detection scheme.
During research, seven growing media, four DNA extraction methods and two PCR protocols as well as two inoculation methods of rhododendron leaves were studied.
It was found that inoculation of plant material in a moist chamber was the most efficient method for maintaining virulence of P.ramorum. Out of the seven media used in the study, V8-2 medium proved to be the most suitable for growing P. ramorum, as fungal sporangia formed best in it. Morphology of P.ramorum mycelium corresponded to that described in literature on PARP/H, carrot piece, V8-1, V8-3 and V8-4 media, whereas it was morphologically different - very thin and non-coralloid on V8-2 media. The most significant structures for identification - chlamydospores and sporangia were morphologically similar in all media.
Method described by Llop et al. (1999) was the most suitable for P.ramorum identification in plant material. Llop et al. (1999) and Qiagen DNeasy kits for plant DNA extraction were the most suitable as they allowed better quality DNA to be obtained, in comparison to the other methods studied during the research. DNA extracted by Llop et al. (1999) and Qiagen DNeasy kits showed similar results in conventional and nested PCR.
