HCV CTL epitopu saturošo rekombinanto proteīnu konstruēšana, attīrīšana un raksturošana

Bakalaura darba mērķis bija konstruēt C hepatīta vīrusa T citotoksisko limfocītu epitopu saturošus rekombinantus proteīnus, raksturot tos un veikt iegūto vīrusiem līdzīgo daļiņu attīrīšanu.
Darbam tika izmantotas nemodificēta (wild type) un modificēta B hepatīta vīrusa core antigēna saturošas ekspresijas plazmīdas, kurās tika klonēts CHV NS3 proteīna 1436-1444 as fragments, izveidojot divas rekombinantas konstrukcijas – ZK1 un ZK2. Rekombinantie proteīni tika ekspresēti E.coli šūnās, attīrīti un raksturoti. Rekombinatie proteīni ir šķīstoši un veido vīrusiem līdzīgas daļiņas. Ekspresijas līmenis ZK2 rekombinantai konstrukcijai (modificēts HBcAg) ir lielāks par ZK1 (nemodificēts HBcAg) taču vīrusiem līdzīgās daļiņas efektīvāk veidojas ZK1 konstrukcijas gadījumā.
Atslēgvārdi: C hepatīta vīruss, CTL epitopi, HBcAg, rekombinanti proteīni, VLD. The aim of the study was to construct the recombinant proteins containing HCV cytotoxic T lymphocytes epitope, to characterize them and to purify the produced virus like particles.
Two E.coli plasmids expressing wild type and modified hepatitis B virus core antigen where used in the study. HCV NS3 protein 1436-1444 aa fragment was inserted into HBcAg creating new recombinant constructs – ZK1 and ZK2. Examination of recombinant protein’s expression and characterization and purification of expression’s products were performed. Recombinant proteins were synthesized, it was found that they are soluble and form viral like particles. The level of expression of ZK2 recombinant construct based on modified HBcAg was higher than of ZK1 based on wild type HBcAg, but recombinant construct based on wild type HBcAg produces virus like particles more efficient.
